兔瘟病毒西宁分离株的分子鉴定及进化分析

【摘要】 本研究首次对处于青藏高原的西宁的兔瘟病毒分离株(命名为RHDV-QHXN1)进行了VP60基因克隆、测序,并与参考株、我国各地区流行株和其他国家流行株进行了基因非同义置换氨基酸分析,以及基因同源性的比较和进化树分析。兔子解剖病理学显示为典型的兔瘟临床症状;并进行了PCR分子鉴定,获得了预期大小的基因片段,测序分析比对分析后确定为兔瘟病毒,命名为RHDV-QHXN1分离株。该分离株与其他参考毒株AY269825(中国江苏南京)和DQ530363(中国陕西杨凌)的核苷酸同源性分别为97.41%和97.13%,氨基酸同源性达99.14和99.31%。进化树分析显示,RHDV-QHXN1与AY269825(中国江苏南京)株聚成微小分支。对RHDV-QHXN1分离株VP60基因的非同义置换的氨基酸分析结果显示,突变位点共有6处,分别位于第351位、359位、370位、432位、508位和573位的氨基酸处。

【Abstract】 In the present study,the Qing Hai Xi Ning isolate of Rabbit viral Hemorrhagic Disease Virus(named RHDV-QHXN1) was identified by the molecular method,and the VP60 gene of it was cloned and sequenced for the first time in the Qinghai-Tibet Plateau Area. The non-synonymous amino acid mutation analysis,gene homology comparison and phylogenetic tree analysis were performed with reference strains and epidemic strains from the various regions of China and other countries. The rabbit anatomical pathology showed that it was typical clinical symptoms of rabbit viral hemorrhagic disease,the PCR molecular identification was conducted and the expected size gene fragment was obtained,after the sequencing and comparison analysis,it was determined as Rabbit viral Hemorrhagic Disease Virus,named RHDV-QHXN1 isolate. The nucleotide homologies of the VP60 of the isolate with other reference strains AY269825(Nanjing,Jiangsu,China) and DQ530363(Yangling,Shaanxi,China) were 97. 41% and 97. 13%,respectively,and the amino acid homologies were 99. 14 and 99. 31%. The phylogenetic analysis showed that RHDV-QHXN1 and AY269825(Nanjing,Jiangsu,China) were clustered into tiny branch. The non-synonymous amino acid mutation analysis of the VP60 gene of the RHDV-QHXN1 isolate revealed that there were six mutation sites at amino acid positions 351,359,370,432,508 and 573,respectively.
