人外周血淋巴细胞染色体G显带标本制备方法探讨
Sample preparation method of chromosome G banding in peripheral blood lymphocytes
【摘要】 目的探讨提高人外周血淋巴细胞染色体G显带效果的方法。方法人外周血收集、淋巴细胞培养及染色体标本制备、Giemsa染色、G显带等操作步骤同常规方法,按不同的烤片温度和时间分组,对比各组标本的显带效果。结果 80℃烤片1~2 h,60℃1~2 h,60℃3~4 h时G显带带纹清晰,效果比较好;而60℃30 min,80℃30 min,80℃3~4 h时显带效果差。结论烤片温度、时间长短直接影响G显带标本的质量,适宜的温度、时间能达到良好显带效果,甚至可提高临床对染色体病的诊断效率。
【Abstract】 Objective To explore the method of improving chromosome G banding in human peripheral blood lymphocytes.Methods Human peripheral blood collection,lymphocyte culture and chromosome specimen preparation,Giemsa staining and G banding procedure with conventional methods,such as grouping according to the different baking temperature and time,contrast each specimen banding effect.Results 80℃ roasting 1 ~ 2 h,60℃ 1 ~ 2 h,60℃ 3 ~ 4 h,the G-banding band is clear,the effect is better. 60℃ 30 min,80℃ 30 min,80℃3 ~ 4 h banding effect is poor. Conclusions The roasting temperature and time directly influence the banding quality,appropriate roasting temperature and time could achieve a good banding effect,and even improve the clinical diagnostic efficiency of chromosomal diseases.
