睡眠剥夺对SD大鼠体重和脏器系数的影响

【摘要】 目的探讨短期完全睡眠剥夺对大鼠体重和脏器系数的影响。方法 SD大鼠16只,雌雄各半,随机分入实验组和对照组,对照组饲养于不锈钢笼具中,实验组饲养于小平台睡眠剥夺装置中,持续7天。于实验开始和结束时称量大鼠体重,于结束时麻醉处死大鼠,解剖脏器并称重,计算脏器系数。结果急性完全睡眠剥夺后,实验组大鼠体重减轻,对照组大鼠体重增加,但实验前后组间和组内比较,差异均无统计学意义。实验组脾脏的脏器系数显著低于对照组,肺和心脏的脏器系数显著高于对照组。两组胸腺、肝脏和肾脏的脏器系数差异无统计学意义。睡眠剥夺大鼠心肌细胞出现明显的溶解现象,细胞间隙增大,血管腔有栓塞行成。结论短期完全睡眠剥夺可致大鼠体重减轻,脾脏、肺和心脏的脏器系数降低,提示短期完全睡眠剥夺对免疫系统、呼吸系统和心血管系统有损害作用,具体损伤及机制有待进一步研究。

【Abstract】 Objective To investigate the short-term total sleep deprivation on health damage in rats.Methods 16 SD rats,male( n = 8) and female( n = 8),were randomly divided into experimental group and control group,the control group was housed in stainless steel cages and the experimental group was housed on a small platform in sleep deprivation device,lasts for 7 days. At the beginning and at the end of the experiment,the rats were weighed. At the end of the experiment,the rats were anesthetize and sacrificed and the organs were dissected and weighing,calculate the organ coefficient.Results After acute sleep deprivation,body weight of the rats in the experimental group decreased,on the contrary rats body weight in control group increased,but before and after the experiment,the differences within groups and among groups were not statistically significant. The organ coefficient of spleen in the experimental group was significantly lower than the control group,the lung and heart organ coefficient were significantly higher than the control group. The two groups had no statistically significant differences in thymus, liver and kidney organ coefficient. In the sleep deprivation group, the myocardial cells showed obvious dissolution,the intercellular space increased,and the blood vessel cavity was embolized. Conclusions The short-term total sleep deprivation leads to the decrease of body mass,as well as the organ coefficient of spleen,lung,and heart. The results demonstrated that short-term total sleep deprivation has damage on rat immune system,respiratory system and the cardiovascular system,however,the specific injury and the mechanism need to be further studied.
