目的通过检测Med19在舌癌组织表达的基础上,探讨Med19表达下调对Tca8113细胞增殖的影响。方法应用免疫组织化学染色方法检测Med19在舌鳞癌组织及舌乳头状瘤组织中的表达,分析Med19的表达分布及表达水平;构建Med19 si RNA并转染人舌鳞癌细胞Tca8113,应用MTT检测细胞增殖的改变。结果免疫组织化学染色结果显示Med19在舌癌组织中高表达;建立si RNA-Med19重组慢病毒载体并转染Tca8113,MTT结果显示Med19下调可以抑制细胞增殖。结论 Med19在舌鳞状细胞癌中高表达;通过si RNA-Med19慢病毒载体转染Tca8113细胞可以抑制细胞增殖,为后续体内实验奠定坚实的基础。

Objective On the basis of the expression of Med19 tongue carcinoma, this study was to investigate the effect of Med19 gene on the proliferation rate of Tca8113 cells. Methods Immunohistochemistry was used to detect the distribution and the expression level of Med19 in tongue carcinoma. Med19 was knocked down in Tca8113 by transfection with the recombinant lentivirus.
