低温混合菌的构建及其降解效能研究

【摘要】 从山东省济宁市南四湖人工湿地中取样,在(4.5±1)℃条件下对样本进行低温驯化和富集培养.经分离、纯化后,最终筛选出5株具高效降解性能的耐冷菌.然后进行不同组合,将其应用于模拟污水处理工艺中,进行协调性试验,并测定每种组合对氨氮、总磷、COD的去除率.通过择优选择,得到污水降解效果最好的一组低温细菌组合1,4,5,编号为TM-1,TM-2和TM-3.通过菌株形态学观察、生理生化特征研究,以及16SrDNA序列分析,对菌株TM-1,TM-2和TM-3进行初步鉴定,并就其生长特性进行了探讨.经鉴定,菌株TM-1,TM-2和TM-3分别为冷杆菌属(Psychrobacter)、鞘脂杆菌属(Sphingobacterium)、假单胞菌属(Pseudomonas).当pH约为6.0,以蔗糖为碳源,以蛋白胨为氮源时,菌株TM-1,TM-2和TM-3达到最适生长条件.通过模拟废水实验测定,48h内混合菌株对氨氮、总磷、COD的去除效率分别可达65.95%,55.43%,65.5%.结果表明,该混合菌株在低温条件下能较好地生长,且对污水中多种常规污染物均表现出稳定的降解特性,对于改善我国北方地区冬季低温下人工湿地的污水处理效率具有一定的潜在价值.

【Abstract】 Collected from Jining City,Shandong Province,Nansi Lake Constructed Wetland,the samples were subjected to low temperature acclimation and enrichment culture at(4.5±1)℃.After isolation and purification,5 strains of psychrotrophic bacteria were finally screened out.Then these five bacteria were combined in various ways and then released in simulated sewage treatment process for further coordination test,in which the removal rates of ammonia nitrogen(NH3-N),total phosphorus(TP)and COD were determined of each combination.Consequently,the combination with three specific strains(1,4 and5)of Psychrotrophic bacteria,which had the highest treatment efficiency,were selected,and these strains were named TM-1,TM-2 and TM-3 respectively.In addition,the preliminary identification was carried out based on the morphological observation,physiological and biochemical characteristics as well as 16 S rDNA sequence analysis of the strains.Beside the above-mentioned details,the growth condition of the strain TM-1,TM-2 and TM-3 were also investigated.Results indicated that strain TM-1,TM-2 and TM-3 were Psychrobacter,Sphingobacterium and Pseudomonas respectively.The strains reach maximal growth rate at optimal growth conditions with pH value approximately 6.0,sucrose as carbon source and peptone as nitrogen source respectively.In the simulated sewage experiment,the removal efficiencies for ammonia nitrogen,total phosphorus and COD by the mixed bacteria strain reached 65.95%,55.43%,65.5%respectively.The results indicated that this group of mixed bacteria strain grew well the wnder simulated low temperature conditions and showed stuble degradation characteristlcs to several conventional pollutants in the sewcse.It appeared that the mixed bacteria strain had a considerable potential value in solving the poor sewage treatment efficiency of constructed wetland in the low-temperature conditions during winter in China’s northern areas.
