一株抑黄曲霉细菌的分离鉴定及培养条件的优化
Screening and Identifying of Inhibition of A.flavus Strain and Response Surface Optimization of Culture Conditions
【摘要】 从长期被黄曲霉污染的土壤中筛选到一株对黄曲霉(Aspergillus flavus 3950)具有极高抑制作用的细菌,对菌株进行形态观察及分子鉴定,确定其为芽孢杆菌属,与Bacillus mojavensis isolate 6(FJ907189.1)亲缘关系最近,且相似度达到99%,命名为Bacillus mojavensis W-1(登录号:KX254606).利用响应面法优化培养基,确定最佳碳源为5%蔗糖,氮源为2.5%胰蛋白胨,培养基最佳pH值为7,优化后的抑菌实验条件下,Bacillus mojavensis W-1发酵上清液对黄曲霉3950的抑菌圈直径从1.6cm提高到2.5cm,优化效果明显.
【Abstract】 It was selected from the soil contaminated by A.flavus in the long time,and the bacteria that had extremely high inhibitory effect on aspergillus flavus were observed and identified by the morphological observation and molecular identification of the strain,and the bacteria was identified as bacillus.According to morphological observation and molecular identification,the selected strain’s relationship was the closest to the Bacillus mojavensis isolate 6,which was nominated as Bacillus mojavensis W-1.The best culture condition was confirmed by response surface method:5% sucrose,2.5% tryptone,and the most appropriate pH was 7.The antibacterial circle enlarged from 1.6 cm into 2.5 cm in the optimum culture condition.
