钴纳米粒子直接化学发光体系检测力竭运动小鼠心肌连接蛋白43含量
Determination of Connexin 43 in Rats After Exhaustive Exercise Based on Cobalt Nanoparticles Labeling
【摘要】 以磁性纳米粒子为载体,利用抗原抗体识别作用将探针捕获在磁珠表面,用钴纳米粒子标记抗体为探针,利用鲁米诺-CoNPs-H2O2直接化学发光体系检测化学发光信号强度,根据化学发光信号强弱实现心肌连接蛋白43含量的测定。心肌连接蛋白43的质量浓度在0.07~20ng·mL-1时,方法检测限为0.03ng·mL-1。并利用该方法对力竭运动后小鼠心肌连接蛋白43含量进行了测定。
【Abstract】 A new chemiluminescence method for connexin 43 detection was developed based on cobalt nanoparticles labeling.Firstly,the cobalt nanoparticles labeled antibody was prepared and the connexin 43 antibody was immobilized onto the surface of magnetic bead.Through the recognition of antigens-antibody the cobalt labeled abtibody was captured onto the magnetic bead in the presence of connexin 43.Under the optimal experimental condition,the concentration of connexin 43 is linear with electrochemical signals in the range from0.07—20 ng·mL-1,the detection limit is 0.03 ng·mL-1.The method was successfully applied to the determination of connexin 43 in muscle of rats after exhaustive.
