高脂饮食诱导C57BL/6小鼠睾丸微血管损伤导致非感染性睾丸炎症反应
High fat diet-induced impairment of testicular microvasculature in C57BL/6 mice leads to non-infectious testicular inflammation
【摘要】 目的探讨高脂饮食诱导雄性C57BL/6小鼠睾丸微血管损伤与高脂诱导的睾丸非感染性炎症反应的关系。方法雄性C57BL/6小鼠,完全随机法分为高脂组(high fat diet,HFD,n=20)和对照组(control,n=20)。对照组普通喂养,高脂组小鼠高脂饮食喂养,共喂养20周,每周检测体质量及血糖。激光多普勒血流灌注监测系统检测两组小鼠睾丸微血管血流灌注量。HE染色观察生精细胞及睾丸微血管形态学变化;免疫组化法检测睾丸微血管内皮细胞CD31、炎症因子TNFα及巨噬细胞特异性标志物F4/80的表达水平;ELISA法检测血清及睾丸组织匀浆中炎症因子TNFα和MCP-1的含量;TUNEL染色法检测小鼠睾丸生精细胞凋亡水平。结果高脂组小鼠体质量及血糖水平均显著增加与对照组比较,差异具统计学意义(P<0.01);睾丸微血管平均血流灌注水平显著低于对照组(P<0.001);HE染色显示高脂组小鼠成熟生精细胞减少,睾丸间质膨大,结构松散;高脂组睾丸微血管CD31表达低于对照组(P<0.05),TNFα及F4/80表达水平高于对照组(P<0.01);TNFα和MCP-1在血及睾丸蛋白中的表达量显著高于对照组(P<0.001);TUNEL染色结果显示生精细胞凋亡水平高于对照组(P<0.01)。结论高脂饮食诱导C57BL/6小鼠血糖增高致睾丸微血管损伤并发非感染性睾丸炎症反应。
【Abstract】 Objective To investigate the relationship between microvascular injury and testicular noninfectious inflammatory response induced by high fat diet in male C57 BL/6 mice. Methods Male C57 BL/6 mice were randomly divided into high fat diet(HFD) group and the control group. The mice in the control group were fed with normal diet, and the mice in HFD group with high fat diet for 20 weeks. Blood glucose and body weight were measured weekly. Laser Doppler perfusion imaging system was used to detect the microvascular perfusion of the mice. HE staining was used to observe the spermatogenic cells and the microvascular changes of testis. The expressions of CD31, TNFα and F4/80 in testis tissues were detected by immunohistochemistry. The concentrations of TNFα and MCP-1 in serum and testis were detected by ELISA. The apoptosis of spermatogenic cells in testis of mice was detected by TUNEL staining. Results Compared with those of the control group, the body weight and blood glucose of HFD group were significantly increased(P <0.01), but the mean blood perfusion level of testis microvessels was significantly decreased(P <0.001). HE staining analysis showed that the number of spermatogenic cells in testis of mice in the HFD group decreased and testicular interstitium presented inflated, loose structure. The expressions of TNFα and MCP-1 in the HFD group were remarkably higher than those in the control group(P <0.001). The concentrations of TNFα and F4/80 in the blood and testicular protein were significantly higher than those in the control group(P <0.01), CD31 in the HFD group was decreased(P<0.05). TUNEL staining analysis showed a significant increase in apoptosis of spermatogenic cells in the HFD group than that in the control group(P <0.01). Conclusion High fat diet-induced hyperglycemia in C57 BL/6 mice leads to testicular microvascular injury complicated with non-infectious testicular inflammation.
