TMCO1蛋白2种不同亚型的表达分析
The Expression Analysis of Two Different Isoforms of TMCO1
【摘要】 内质网钙离子通道蛋白TMCO1具有2个不同的亚型,分别表达239个氨基酸和188个氨基酸,而2种亚型在细胞中的表达还缺少研究论证.通过对氨基酸序列的分析,比较了2种不同亚型的结构域,并构建得到2种不同亚型的哺乳动物细胞表达载体.免疫荧光实验和蛋白质免疫印迹实验表明了2种蛋白亚型的亚细胞定位和表达,同时表明在哺乳动物细胞中,TMCO1的m RNA更倾向于在下游的起始密码子翻译.进一步通过RNA干扰实验发现敲低TMCO1的表达对细胞周期没有明显的影响.
【Abstract】 ER membrane protein TMCO1 has two different isoforms, which predict to be 239 or 188 amino acids. However, the expression patten is still unknown. Here, compared the predicted transmembrane domains of two different isoforms based on the amino acids alignment, also constructed plasmids of two isoforms. The immunofluorescence assay showed us the same location of two isoforms. The western blot assay indicated the m RNA of TMCO1 prefers to translate from the second ATG. Knockdown assay confirmed that TMCO1 has no impact on cell cycle.
