鳙鱼鱼骨脱钙工艺优化及其模型

【摘要】 为了顺利地从鳙鱼鱼骨中提取较高纯度的胶原蛋白,进行了鳙鱼鱼骨脱钙的预处理研究。采用单因素实验和响应面实验考察了主要工艺参数液固比、盐酸质量分数、搅拌速度和搅拌时间对脱钙率的影响,并对它们进行了优化,得到了最佳参数分别为7.3∶1 g·g-1、7.00%、1 000 r·min-1和1.0 h,在此条件下,脱钙率达到91.68%(实际值)。建立了以考察的4个工艺参数为自变量,以脱钙率为响应值的工艺模型,该模型理论值与实验值吻合度较高,可预测不同工艺条件下的脱钙率。同时,确定和比较了各工艺参数对脱钙率影响的显著性程度,为严格控制操作指标提供了理论依据。

【Abstract】 The pretreatment study of decalcification of bighead carp fish bone was carried out in order to fluently extracting purer collagen from it. Single experimental method and response surface experimental method were used to investigate the effects of main process parameters of ratio of liquid to solid,hydrochloric acid mass fraction,stirring speed and stirring time on fish bone decalcification rate. The parameters were optimized to be liquid/solid ratio7. 3∶1 g·g-1,hydrochloric acid mass fraction 7. 0 %,stirring speed 1 000 r·min-1 and stirring time 1. 0 h,respectively,under which the decalcification rate reached up to 91. 78%. The model with the four parameters as independent variables and decalcification rate as response value was established,whose theoretical values have good agreement with experimental values and can be used to predict decalcification rate at different process conditions. The significant degree of effects of process parameters on decalcification rate were determined and compared,which can provide the theoretical basis for strictly controlling the operation indexes.
