淫羊藿苷对IgA肾病大鼠纤维化和炎症反应的调控作用
Icariin regulates the fibrosis and inflammatory reaction in IgA nephropathy rats
【摘要】 目的Ig A肾病(Ig A nephropathy,Ig AN)是最常见的原发性肾小球肾炎。本文旨在探索淫羊藿苷对Ig AN大鼠纤维化和炎症反应的影响。方法牛血清球蛋白(bovine gamma globulin,BGG)处理建立Ig AN大鼠模型。免疫组化和蛋白印迹检测转化生长因子β1(transforming growth factor beta1,TGF-β1),IV胶原蛋白(type IV collagen,Col IV)和纤连蛋白(Fibronectin)1蛋白水平。酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)检测血清白细胞介素1β(interleukin 1β,IL-1β),IL-6和IL-18水平。蛋白印迹分析细胞核中抗肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)、NF-κB P65和抗血管细胞黏附蛋白(vascular cell adhesion protein 1,VCAM-1)表达。结果与对照组相比,Ig AN模型组尿蛋白,血清肌酐和尿素氮水平明显升高(P<0.05)。模型加药组尿蛋白,血清肌酐和尿素氮水平明显低于Ig AN模型组(P<0.05)。Ig AN模型组TGF-β1、Col-IV和Fibronectin 1表达水平显著高于对照组(P<0.01)。淫羊藿苷处理Ig AN模型大鼠后,TGF-β1、Col-IV和Fibronectin 1表达水平明显下降(P<0.05)。而且,Ig AN模型组血清中IL-1β、IL-6和IL-18水平明显高于对照组(P<0.05)。与Ig AN模型组相比,模型加药组IL-1β、IL-6和IL-18水平明显降低(P<0.05)。另外,Ig AN模型大鼠肾脏组织细胞核的NF-κB P65、TNF-α和VCAM-1蛋白水平显著高于对照组(P<0.01)。与Ig AN模型组相比,模型加药组大鼠肾脏组织细胞核的NF-κB P65、TNF-α和VCAM-1蛋白水平明显下降(P<0.05)。结论淫羊藿苷可改善Ig AN大鼠纤维化水平并抑制炎症反应。
【Abstract】 IgA nephropathy(IgAN) is the most common forms of primary glomerulonephritis. This study aims to explore the effect of icariin on fibrosis and inflammatory reaction in IgAN rats. IgAN rat model was induced by treatment of bovine gamma globulin(BGG). Then immunohistochemistry and Western blot were applied to detect the protein levels of transforming growth factor beta1(TGF-β1), type IV collagen(Col IV) and fibronectin 1; the serum levels of interleukin(IL)-1β,IL-6 and IL-18 were detected by enzyme-linked immunosorbent assay(ELISA); while the expression of tumor necrosis factor alpha(TNF-α), NF-κB P65 and vascular cell adhesion protein 1(VCAM-1)in the nucleus was measured by Western blotting. Data showed that the levels of proteinuria, serum creatinine and urea nitrogen were higher in IgAN model group than those in control group(P<0.05), and lower in treatment group than those in IgAN model group(P<0.05). The protein levels of TGF-β1, Col-IV and fibronectin 1 in IgAN model group were higher than control group(P<0.01). After treatment with icariin, the protein levels of TGF-β1, Col-IV and fibronectin 1 in IgAN rats were reduced(P<0.05).Moreover, the serum levels of IL-1β, IL-6 and IL-18 were higher in IgAN model group than those in controlgroup(P<0.05), and lower in treatment group than those in IgAN model group(P<0.05). In addition, the protein levels of nuclear NF-κB P65, TNF-α and VCAM-1 in IgAN were higher in model group than those in control group(P<0.01), and lower in treatment group than those in IgAN model group(P<0.05). Taken together, icariin ameliorates fibrosis and inhibits inflammatory reaction in IgAN rats.
