来自古菌的纤维素酶对棉纤维降解条件的优化

【摘要】 以来自古菌的一种新的纤维素酶作用于新疆长绒棉,对其进行降解分析,从酶解温度、p H、酶解时间及酶用量,通过DNS法,考察了4个因素对还原糖浓度的影响。通过响应面试验分析发现:这种纤维素酶在p H为7.0,酶用量为2.0 m L/g,酶解温度为70℃,酶解时间为2 h时,对新疆长绒棉的降解效果最佳,还原糖浓度最高为0.034 9 mg/m L。

【Abstract】 A new cellulase from archaea was used to degrade Xinjiang long-staple cotton. Through DNS method, the effect of reaction temperature, p H, enzyme dosage, reaction time on cotton degradation were investigated. Through the analysis of response surface test, it was found that the cellulose had the best effect on the Xinjiang long-staple cotton when the p H was 7, the cellulase dosage was 2 m L/g, the reaction temperature was 70 ℃, the reaction time was 2 h, and the highest reducing sugar concentration was 0.034 9 mg/m L.
