不同浓度半胱氨酸对幼龄陶寒杂种羊卵母细胞发育的影响

【摘要】 为了探讨半胱氨酸对幼龄陶寒杂种羔羊卵母细胞发育的影响,试验采用激素诱导4~8周龄陶寒杂种羔羊获得624枚A、B级卵丘卵母细胞复合体(COCs),分别在含不同浓度半胱氨酸的成熟液里成熟培养24 h,对照组(0μmol/L)、试验Ⅰ组(100μmol/L)、试验Ⅱ组(150μmol/L)和试验Ⅲ组(200μmol/L),经体外受精检测卵母细胞发育效果。结果表明:试验Ⅰ组、Ⅱ组和Ⅲ组成熟卵母细胞的卵裂率分别为60.9%、62.82%和64.1%,比对照组的卵裂率59.62%分别高1.28、3.2和4.48个百分点(P>0.05),试验Ⅰ组、Ⅱ组和Ⅲ组成熟卵母细胞的囊胚率分别为21.79%、23.08%和23.72%,比对照组的囊胚率18.59%分别高3.2、4.49和5.13个百分点(P>0.05)。说明成熟液里添加一定量半胱氨酸有利于卵母细胞成熟和发育。

【Abstract】 In order to explore the effect of cysteine on the development of oocytes in young pottery and cold hybrid sheep, 624 A and B grade oocyte complexes(COCs) of 4-8-week-old young pottery and cold hybrid sheep with hormone-induced were tested in groups with different concentrations of cysteine, control group(0 μmol/L), Group Ⅰ(100 μmol/L), Group Ⅱ(150 μmol/L) and Group Ⅲ(200 μmol/L), after 24 h maturation in mature liquid IVF was carried out to investigate the development competence of oocytes. The results showed that the cleavage rates were 60.9%,62.82% and 64.1% in GroupⅠ, GroupⅡ and group Ⅲ respectively, which were 1.28, 3.2 and 4.48%higher than that in the control group(59.62%)(P>0.05). The blastocyst rates of groupⅠ, GroupⅡand group Ⅲ were 21.79%, 23.08% and 23.72% respectively compared with the rate of control group18.59%. It is suggested that the addition of a certain amount of cysteine in the mature liquid is beneficial to the maturation and development of oocyte.
