参松养心胶囊促血管新生的实验研究
Effect of Shensongyangxin capsule on angiogenesis
【摘要】 目的观察参松养心胶囊对鸡胚绒毛尿囊膜(chick embryo chorioallantoic membrane,CAM)血管新生的影响,探讨该药促进血管新生的可能机制。方法随机各取15只雄性Wistar大鼠分别制作常规血清和参松养心胶囊血清。建立CAM模型,将60枚存活鸡胚随机分为生理盐水组、空白血清组、血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)组和参松养心胶囊组,每组15枚。观察血管新生形态,计数各组CAM上载体周围血管数目,体外培养人脐静脉内皮细胞,应用MTT法测定各组吸光度(A)值。结果与生理盐水组比较,VEGF组和参松养心胶囊组血管以载体为中心呈辐辏状生长,血管总数明显增多,有显著差异[(33.71±3.36)支和(34.51±3.34)支vs(22.11±2.05)支,P<0.05]。与生理盐水组和空白血清组比较,参松养心胶囊组A值明显增加,差异有统计学意义(1.23±0.21 vs 0.91±0.15和0.96±0.14,P<0.05)。结论参松养心胶囊具有促进CAM血管新生作用,其机制可能与促进内皮细胞增殖有关。
【Abstract】 Objective To study the effect of Shensongyangxin capsule on angiogenesis in chick embryo chorioallantoic membrane(CAM)and its mechanism.Methods Fifteen male Wistar rats were randomly selected for the parathion of conventional serum and Shensongyangxin capsule serum.After a chick embryo CAM model was established,60 surviving chicken embryos were randomly divided into normal saline group,blank serum group,vascular endothelial growth factor(VEGF)group and Shensongyangxin capsule group(15 in each group).The morphology of angiogenesis was observed and the number of vessels around the carrier of CAM vessels was counted in different groups.The human umbilical vein endothelial cells were cultured in vitro.The absorbance value in different groups was measured with MTT staining.Results The blood vessels grew in a convergent manner with the carriers of CAM vessels as their center.The total number of blood vessels was significantly greater in VEGF group and Shensongyangxin capsule group than in normal saline group(33.71±3.36 and 34.51±3.34 vs 22.11±2.05,P<0.05).The absorbance value was significantly higher in Shensongyangxin capsule group than in normal saline group and blank serum group(1.23±0.21 vs 0.91±0.15 and 0.96±0.14,P<0.05).Conclusion Shensongyangxin capsule plays a role in promoting angiogenesis of click embryo CAM by increasing the proliferation of endothelial cells.
