采用大孔吸附树脂法分离纯化金花葵茎叶总黄酮的研究
STUDY ON SEPARATION AND PURIFICATION OF TOTAL FLAVONOIDS FROM STEM & LEAF OF Hibiscus manihot BY MACROPOROUS ADSORPTION
【摘要】 为了对金花葵进行全面的开发利用,研究分离纯化金花葵茎叶中总黄酮的最佳条件,选择10种大孔树脂,以金花葵茎叶中总黄酮的含量为参考指标,筛选出其中最优树脂,并对其树脂的最佳吸附及解吸参数进行探究,结果表明,在样液浓度为3mg/mL,pH值为5.0,吸附流速为3BV/h,过柱次数为3次,70%乙醇的洗脱速率为4BV/h的条件下,LX-J24大孔树脂对总黄酮的吸附与解吸效果均为最佳,经验证金花葵茎叶中总黄酮的含量提取率为78.06%,纯度高达57%,即采用LX-J24大孔树脂对金花葵茎叶总黄酮进行分离纯化是较为理想的,为金花葵的综合利用提供了参考依据。
【Abstract】 In order to study and optimize the total flavonoids in the stems and leaves of Hibiscus manihot, 10 kinds of macroporous resins were selected to study the content of total flavonoids in the leaves of Hibiscus manihot as reference index. The optimum adsorption and desorption parameters of the resin were investigated. The results showed that when the concentration of the sample was 3 mg/mL, the pH was 5.0, the adsorption flow rate was 3 BV/h. The effect of LX-J24 macroporous resin on the adsorption and desorption of total flavonoids was the best under the conditions of 3 times and the elution rate of 70% ethanol was 4 BV/h. The extraction rate of flavonoids was 78.06% and the purity was up to 89%. It was ideal to separate and purify the total flavonoids from the leaves of Hibiscus manihot with LX-J24 macroporous resin, which provided the reference for the comprehensive utilization of Hibiscus manihot.
