脂多糖对狼疮鼠骨髓幼红细胞巨噬细胞连接蛋白的影响
The effects of lipopolysaccharide on expression levels of erythroblast macrophage protein in a mouse model of lupus
【摘要】 目的:观察脂多糖(LPS)对狼疮鼠骨髓幼红细胞巨噬细胞连接蛋白(EMP)的影响。方法 :将狼疮鼠组(24只)分4组,分别腹腔注射不同浓度LPS(0、2、4及8 mg/kg);正常对照组(24只)分组和LPS处理方法同狼疮鼠组。采用定量聚合酶链式反应(PCR)检测骨髓EMP基因表达,免疫组化法观察骨髓中EMP阳性细胞。结果:(1)相同剂量LPS狼疮鼠组和正常对照组间比较:狼疮鼠组EMP m RNA表达比正常对照组显著性增高;(2)不同剂量LPS狼疮鼠组间比较:8 mg/kg LPS狼疮鼠组EMP m RNA表达高于2 mg/kg和0 mg/kg LPS狼疮鼠组;4 mg/kg和2 mg/kg LPS狼疮鼠组EMP表达较0 mg/kg LPS狼疮鼠组显著性增高;(3)不同剂量LPS正常对照组间比较:8 mg/kg LPS组EMP m RNA表达显著高于0 mg/kg LPS组;(4)免疫组化示EMP在狼疮鼠组及正常对照组中均有表达,细胞核、细胞质和细胞膜均被染色,以细胞质及细胞膜染色明显,随着LPS浓度增高,狼疮鼠组及正常对照组EMP阳性细胞更加密集。结论:EMP可能参与SLE发病,并与狼疮活动程度密切相关。
【Abstract】 Objective: To evaluate the effects of lipopolysaccharide(LPS) on expression levels of erythroblast macrophage protein(EMP) in a mouse model of lupus. Method: Twenty-four lupus mice were divided into 4 groups, and intraperitoneally injected with various concentrations of LPS(0 mg/kg, 2 mg/kg, 4 mg/kg and 8 mg/kg). LPS-treated normal mice served as controls. Expression levels of EMP m RNA in bone marrow were assessed using quantitative PCR while densities of EMP positive cells were evaluated using immunohistochemical staining. Result:(1)Following LPS treatment, expression levels of EMP m RNA were significantly higher in lupus mice than in controls.(2)In lupus mice, expression levels of EMP m RNA were significantly higher in mice treated with 8 mg/kg of LPS than in that treated with either 2 mg/kg of LPS or vehicle. In addition, both 4 mg/kg and 2 mg/kg of LPS significantly elevated expression levels of EMP m RNA in comparison with vehicle.(3)In comparison with vehicle treatment, 8 mg/kg of LPS also significantly increased expression levels of EMP m RNA in normal mice.(4)Immunostaining results showed that EMP mainly expressed in the cytoplasm and cell membrane, but slightly expressed in cell nuclei, of both lupus and control mice. The densities of EMP positive cells in bone marrow were positively correlated with LPS concentrations. Conclusions: EMP could play a pathogenic role in SLE, and its expression is associated with the activity of lupus.
