咪喹莫特诱导的银屑病样小鼠皮肤抗细菌感染能力的研究
A study of the bacterial resistance in imiquimod-treated mice
【摘要】 目的:观察咪喹莫特诱导的银屑病样小鼠皮肤抵抗细菌感染的能力。方法:Balb/c小鼠,背部脱毛后涂抹5%咪喹莫特乳膏,制作银屑病样小鼠模型。取银屑病样小鼠及正常小鼠各10只,在其背部皮内分别注射1×1011菌落形成单位(CFU)/L、1×1012CFU/L金黄色葡萄球菌液及链球菌菌液各40μL,观察皮肤的红斑、脓肿等炎症反应程度,行组织病理检查及取金黄色葡萄球菌脓肿检测组织荷菌量。结果:1×1011 CFU/L金黄色葡萄球菌菌液在正常小鼠皮肤造成直径38 mm的脓肿,银屑病样小鼠则无明显改变;1×1012 CFU/L金黄色葡萄球菌菌液在正常小鼠皮肤造成直径1013 mm的巨大脓肿,在银屑病样小鼠皮肤造成直径25 mm的结痂;1×1011 CFU/L链球菌菌液在正常小鼠皮肤引发红色小丘疹,银屑病样小鼠无反应;1×1012 CFU/L链球菌菌液导致正常小鼠皮肤出现小脓点及结痂,47 d愈合,银屑病样小鼠皮肤出现红色小丘疹,持续存在,并逐渐隆起。银屑病样小鼠皮肤的金黄色葡萄球菌脓肿的荷菌量显著少于正常小鼠。结论:咪喹莫特诱导的小鼠银屑病样皮损对金黄色葡萄球菌及链球菌急性感染的抵抗力显著增强,对链球菌存在迟发的炎症反应。
【Abstract】 Objective: To study the bacterial resistance of psoriasis-like mice induced by imiquimod. Methods: After removal of hair, 5% imiquimod cream was applied topically to the back of Balb/c mice for 7 days to establish psoriasis model.Afterward, both imiquimod-treated mice and controls, 10 mice each, were intracutaneously injected with 40 μL of Staphylococcus aureus and group A hemolytic Streptococcus into separate sites of the back skin at the concentrations of 1×1011 CFU/L and 1×1012 CFU/L. In addition to evaluation of cutaneous inflammatory response, abscesses infected by Streptococcus were removed and grinded to assess the bacterial load. Results: Injection of 1×1011 CFU/L of Staphylococcus aureus induced abscesses sized 38 mm in diameter in normal mice, while in imiquimod-treated mice, the skin appeared normal. Injection of 1×1012 CFU/L of Staphylococcus aureus resulted in huge abscesses sized 1013 mm in diameter in normal mice. In contrast, 1 ×1012 CFU/L of Staphylococcus aureus only caused scabs sized 25 mm in length in imiquimod-treated mice. 1×1011 CFU/L hemolytic Streptococcus caused small red papules in normal mice, while the skin of imiquimod-treated mice remained unchanged. 1×1012 CFU/L Streptococcus induced small pustules or scabs, which resolved in 47 days in normal mice. But in imiquimod-treated mice, small papules gradually enlarged. The Staphylococcus aureus load of imiquimod mice was significantly less than that of normal mice. Conclusions: Treatments with imiquimod increase cutaneous resistance to acute infections of Staphylococcus aureus and Streptococcus haemolyticus. Imiquimod-treated mice exhibit delayed inflammatory response to infections of Streptococcus haemolyticus.
