动/静脉灌注紫杉醇对兔舌VX-2鳞癌的疗效研究
Therapeutic efficacy of paclitaxel via arterial or venous perfusion in rabbits with VX-2 tongue cancer
【摘要】 目的:探讨采用不同给药途径灌注紫杉醇对兔舌VX-2鳞癌的疗效。方法:建立兔舌VX-2鳞癌模型并分为6组,即经舌动脉或耳缘静脉分别灌注紫杉醇脂质体、游离紫杉醇和5%葡萄糖溶液。当颈部转移淋巴结直径> 5 mm时开始给予化疗。7 d后,对原发性灶和转移淋巴结进行细胞学和免疫组化检测VX-2肿瘤细胞凋亡和P53蛋白表达。结果:靶动脉灌注紫杉醇脂质体能有效诱导兔舌VX-2鳞癌原发灶和颈转移淋巴结肿瘤细胞凋亡、抑制增殖。结论:同一给药途径紫杉醇脂质体诱导淋巴结转移癌细胞凋亡较游离紫杉醇强,区域性动脉灌注较全身静脉灌注可更有效诱导淋巴结转移癌细胞凋亡。
【Abstract】 Objective: To investigate the therapeutic efficacy of paclitaxel in different ways in rabbits with VX-2 tongue cancer. Methods: Rabbit VX-2 tongue cancer model was established and animals were then divided into 6 groups,in which animals received perfusion with paclitaxel liposome,with free paclitaxel,or 5% glucose via the lingual artery,with paclitaxel liposome,with free paclitaxel,or 5% glucose via the ear vein independently. When the maximum diameter of cervical lymph nodes was larger than 5 mm,chemotherapy was initiated. Seven days later,flow cytometry and immunohistochemistry were performed to detect the apoptosis of VX-2 cells and P53 expression in the primary cancer and metastatic lymph nodes.Results: Targeted arterial perfusion with paclitaxel liposome was more effective to induce the apoptosis of cancer cells in the primary cancer and metastatic lymph nodes and inhibit their proliferation. Conclusion: Targeted arterial perfusion with paclitaxel liposome is effective to reduce tumor size,attenuate the surgery induced injury and improve the post-operative quality of life of oral cancer patients.
