酸度对MCF-7细胞电化学行为的影响
The effects of acidity on electrochemical behavior of MCF-7 cells
【摘要】 目的:研究不同酸度条件下混合标准品(黄嘌呤、鸟嘌呤、腺嘌呤和次黄嘌呤)的电化学行为,为细胞裂解液电化学响应信号的归属提供依据。方法:采用电化学法对混合标准品及细胞裂解液进行单线扫描,对比分析细胞裂解液电化学信号的归属,同时对比高效液相色谱法检测结果,佐证电化学方法的正确性;进一步检测不同酸度条件下混合标准品电化学响应信号的变化,分析酸度对电化学法检测结果的影响。结果:混合标准品溶液和细胞裂解液分别在0.68V和1.00V左右出现了两个较大的响应信号,可以归属为黄嘌呤和鸟嘌呤、腺嘌呤和次黄嘌呤的混合峰。在pH=7.4、pH=7.2和pH=7.0的缓冲溶液中,黄嘌呤和鸟嘌呤混合峰出峰位置从0.67V处逐渐移动到0.71V处,腺嘌呤和次黄嘌呤混合峰出峰位置从1.00V处逐渐移动到1.03V处。结论:随着pH值的降低,标准品的电化学响应信号不断向高电位方向移动,混合标准品与细胞裂解液电化学响应信号位置的微小差别在于其酸度的差异。
【Abstract】 Objective:To investigate the electrochemical behavior of mixed standards( xanthine,guanine,adenine and hypoxanthine) in different acidity conditions on the purpose of providing the basis for the attribution of electrochemical response of cell suspensions.Methods:The mixed standards and the cell suspensions were detected by the electrochemical method with linearly scan.The correctness of electrochemical method needs confirmation by comparative analysis of the attribution of electrochemical response of cell suspensions and by comparison with the results of high performance liquid chromatography.The electrochemical response of mixed standards( xanthine,guanine,adenine and hypoxanthine) was detected in different acidity conditions to analyze the effect of acidity on the electrochemical detection results.Results:There were two large response signals at 0.68 V and 1.00 V in the mixed standard solution and cell suspensions,which could be attributed to the mixed peak of xanthine and guanine,adenine and hypoxanthine,respectively.In buffer solution with pH = 7.4,pH = 7.2 and pH = 7.0,the peak position of xanthine and guanine was shifted from 0.67 V to 0.71 V,and that of adenine and hypoxanthine was shifted from 1.00 V to 1.03 V.Conclusions:With the decrease of pH value,the electrochemical response signal of the standard should move to high potential direction.The difference of the electrochemical response signal between the mixed standard and the cell suspensions depends on the difference of their acidity.
