小檗碱通过激活线粒体途径诱导口腔鳞癌HN-4细胞凋亡

【摘要】 目的:探讨小檗碱对人口腔鳞癌细胞系HN-4增殖和凋亡的影响及其分子机制。方法:使用不同浓度小檗碱对HN-4细胞进行干预,采用MTS法检测小檗碱对HN-4细胞活性的影响,Annexin-V-FITC/PI双标染色法分析小檗碱对HN-4细胞凋亡率的影响,流式细胞术检测小檗碱对HN-4细胞线粒体膜电位的影响,利用Western Blot法检测小檗碱对HN-4细胞凋亡相关蛋白Bax、Bcl-2、细胞色素C、剪切的半胱氨酸的天冬氨酸蛋白水解酶(c-Caspase)3、c-Caspase 9表达的影响。结果:小檗碱可抑制HN-4细胞的活性,并可诱导其凋亡和线粒体膜电位的减少;经小檗碱干预后,HN-4细胞中凋亡蛋白Bax、细胞色素C、c-Caspase 3和c-Caspase 9蛋白的表达显著上调,而抗凋亡蛋白Bcl-2的表达显著下调。结论:小檗碱可通过激活线粒体凋亡相关信号通路显著抑制人口腔鳞状癌HN-4细胞增殖活性,并诱导其凋亡。

【Abstract】 Objective: To investigate the effect of berberine on the cell viability and apoptosis in human oral squamous cell carcinoma cell line HN-4,and explore the possible molecular mechanisms. Methods: The cultured HN-4 cells were treated with different concentrations of berberine for 24 or 48 h; then detected by MTS method to measure its viability. The apoptosis rate was quantified by using Annexin V-FITC apoptoticdetection kit. The mitochondrial membrane potential changes were detected with Rhodamine 123 dye by flow cytometric assay. The expression of Bax,Bcl-2,Cytochrome C,cleaved-Caspase 3 and cleaved-Caspase 9 was detected by Western Blot. Results: Berberine significantly inhibited the viability of HN-4 cells in a dose-dependent manner. Berberine significantly increased the apoptosis rate and induced theloss of mitochondrial membrane potential on HN-4 cells.Berberine significantly upregulated the expression of pro-apoptotic proteins including Bax,Cytochrome C,cleaved-Caspase 3 and cleaved-Caspase 9,as well as upregulated the expression of anti-apoptotic Bcl-2 protein. Conclusions: Berberine significantlyinhibited the viability and induced apoptosis of HN-4 cells through activating mitochondrial-mediated pathway.
