车前草干、鲜品提取物有效成分含量的对比研究

【摘要】 目的采用相同的提取工艺,比较车前草鲜品和干品提取物中总黄酮和熊果酸的含量差异。方法采用高效液相色谱法测定熊果酸,以Kromasil C18(4.6mm×250mm,5μm)为色谱柱,甲醇:乙腈:0.2%磷酸水(40:50:10)为流动相,流速1ml·min-1,检测波长210nm,柱温25℃。以60%乙醇为提取剂,以芦丁为对照品,采用紫外-可见分光光度法测定总黄酮。结果与车前草干品提取物相比,车前草鲜品提取物的熊果酸含量高出100%以上,总黄酮的含量高出约30%。结论车前草鲜品提取物中熊果酸和总黄酮含量均高于车前草干品。

【Abstract】 Objective To compare the content difference of total flavonoids and ursolic acid sby adopting the same extraction process. Methods An HPLC method was established to determine the content of ursolic acid. Ursolic acid was separated at 25℃on a Kromasil C18(250 mm×4.6 mm,5μm) column with methanol-acetonitrile-0.1% phosphoric acid(40:50:10) as the mobilephase. The detection wavelength was set at 210 nm and the flow rate was 1 ml·min-1. With 60% ethanol as the extraction solvent and rutin as the reference substance. The total flavanone was determined by the Ultraviolet Spectrophotometry. Results The content of ursolic acid in fresh extract was more than 1 time higher than that in the dry extract. And the content of total flavonoids was about 30% higher. Conclusion When the ursolic acid and total flavonoids were used as indicators,the fresh extraction can get better extraction efficiency,which is helpful to improve the utilization efficiency of Plantago asiatica L.
