生物素化微细纤维肽结合IGF-Ⅰ对兔心肌细胞Akt蛋白表达及磷酸化的影响

【摘要】 目的探讨生物素化微细纤维肽结合IGF-Ⅰ后对兔心肌细胞Akt蛋白表达及磷酸化的影响。方法体外培养胎兔心肌细胞,分别以生物素化微细纤维肽结合的IGF-Ⅰ和未结合的IGF-Ⅰ诱导Akt磷酸化,检测Akt、p-Akt蛋白表达,肌钙蛋白表达。结果与未结的IGF-Ⅰ相比,生物素化微细纤维肽结合的IGF-Ⅰ心肌细胞Akt、p-Akt、肌钙蛋白表达水平均显著升高(P<0.05)。结论生物素化微细纤维肽结合的IGF-Ⅰ不但保留了IGF-Ⅰ的生物蛋白活性,并且能加强和延长其作用,增强Akt、p-Akt、肌钙蛋白表达,增加蛋白质合成。

【Abstract】 Objective To study the effect of Biological microfiber peptides combine IGF-Ⅰon the Akt expression and phosphorylation in Rabbit cardiomyocytes. Methods Myocardial cells of rabbits were cultured in vitro,Induced Akt phosphorylation by IGF-Ⅰ,which is combined with biotin microfiber peptide and uncombined,the protein expression of Akt、p-Akt were observed after 24 hours. the protein expression of troponin were observed after 14 days. Results Comparing with uncombined IGF-Ⅰ,the protein expression of Akt,p-Akt troponin were significantly increased in IGF-Ⅰ combined with biotin microfiber peptide Myocardial cells(P<0.05). Conclusion The IGF-Ⅰ combined with biotin microfiber peptide,Not only the IGF-Ⅰ biological protein activity was preserved but also can be enhanced and extended,enhance protein expression of Akt,p-akt and troponin,Increase protein synthesis.
