不同剂量的Anti-Fx1A对大鼠被动型Heymann肾炎模型的影响

【摘要】 目的:通过观察不同剂量的Anti-Fx1A对大鼠被动型Heymann肾炎模型的影响,确定被动型Heymann肾炎模型所需最适Anti-Fx1A剂量。方法:成年雄性SD大鼠分为A、B、C、D 4组,分别经尾静脉注射Anti-Fx1A 4 ml/kg、6 ml/kg、8 ml/kg以及生理盐水8 ml/kg。观察各组大鼠造模成功率,造模前与造模后7 d、14 d、28 d 24 h尿蛋白定量(24 h UP),造模28 d后血白蛋白(Alb)、血清肌酐(Scr)及血三酰甘油(TG),在普通光学显微镜下观察大鼠肾组织的病理学改变,免疫荧光测肾组织Ig G、C3的表达。结果:Anti-Fx1A的剂量与24 h UP及病理学改变可能存在着正相关性,与血白蛋白和造模成功率可能存在着负相关性,与血肌酐变化差异无统计学意义。结论:按大鼠体重6 mg/kg尾静脉注射Anti-Fx1A造模大鼠被动型Heymann肾炎可能是最适剂量。

【Abstract】 Objective: To observe the effect of different doses of Anti-Fx1 A on passive Heymann nephritis model in rats,and determine the optimal anti-Fx1 A dose needed for a passive Heymann nephritis model. Methods: Adult male Sprague-Dawley(SD) rats were randomly divided into four groups and were injected Anti-Fx1 A serum(4 ml/kg,6 ml/kg,8 ml/kg) and physiological saline(8 ml/kg) in different through caudal vein. The success rate of modeling was observed in each group,24 h urinary protein quantification(24 h UP) was measured before and 7 days,14 days and 28 days after modeling. Serum albumin(Alb),serum creatinine(Scr),blood triglyceride(TG) were observed on the 28 th day after modeling,and the pathological changes of rat kidney tissue were observed under ordinary light microscope. Immunofluorescence was used to measure the expression of Ig G and C3. Results: There was a positive correlation between the dose of Anti-Fx1 A and 24 h UP and pathological changes,and there may be a negative correlation between the dose of Anti-Fx1 A and serum albumin and the success rate of modeling. There was no statistically significant difference between the dose of Anti-Fx1 A and serum creatinine. Conclusion: Passive Heymann nephritis in rats modeled by injecting anti-Fx1 A at a rat body weight’s 6 mg/kg may be the optimal dose.
