优化冰冻血小板制备条件的初步研究

【摘要】 目的探讨最佳的冰冻血小板制备条件。方法分组使用两种浓度(75%或100%)的冰冻血小板保护剂二甲基亚砜(DMSO),根据质量体积比加入需冰冻的血小板中,使之终浓度为5%,采用不同解冻方法复苏血小板,对比组间差异。结果 DMSO的浓度、注入速度和操作状况对血小板质量均有一定影响;各种制备条件所得的结果的差异均有统计学意义;DMSO原液与75%DMSO制备的冰冻血小板复融后絮状物的出现差异有统计学意义。结论 75%的DMSO更适合制备冰冻血小板。

【Abstract】 Objective Explore the feasibility for advanced preparation and gathering of frozen platelets. Methods Add DMSO100% and 75% solution to ready-to-freeze platelets respectively until DMSO solution percentage stands at 5%. Different methods were used to resuscitate platelets and compare the results. Results DMSO concentration,speed of DMSO injection and processing conditions all contribute to platelets quality. The results that platelets yielded under each conditions in the process show statistical significance. And the floccules formed in DMSO 100% and 75% solution during the thawing process also show statistical significance. Conclusion DMSO 75% solution is more effective for effective preparation and gathering of frozen platelets.
