高效液相色谱-荧光法测定果蔬及谷物中15种欧盟优先控制的多环芳烃

【摘要】 目的建立一种果蔬及谷物中15种欧盟优先控制的多环芳烃(polycyclic aromatic hydrocarbons,PAHs)的凝胶渗透色谱-高效液相色谱-荧光(GPC-HPLC-FLD)检测法。方法样品经乙腈提取,浓缩近干后,用环己烷-乙酸乙酯混合溶剂复溶,经凝胶渗透色谱净化,高效液相色谱分离,测定各种PAHs在不同激发波长和发射波长处的荧光强度,外标法定量。结果15种PAHs的线性范围为0.1~20μg/mL,线性关系良好,相关系数r>0.999。在3个添加水平下的平均回收率为60.0%~137.8%,RSD<16.0%。检出限为0.1μg/kg。结论该方法选择性强、灵敏度高、处理方法简单,可用于果蔬及谷物中15种PAHs的测定。

【Abstract】 Objective To establish a method for the determination of 15 kinds polycyclic aromatic hydrocarbons(PAHs)controlled by European Union with priority in fruits,vegetables and cereals using gel permeation chromatography-high performance liquid chromatography-fluorescence detector(GPC-HPLC-FLD)method.Methods PAHs were extracted from samples by acetonitrile and the extracts were concentrated to nearly dried,then redissolved in cyclohexane-ethylene acetate,which were purified by GPC and separated by HPLC.The fluorescence intensity of various PAHs at different excitation and emission wavelengths was determined.The external standard method was used for quantification.Results The calibration graphs were linear in the concentration range between 0.1 to 20μg/mL with the correlation coefficients more than 0.999.The average recoveries at the three spiked levels were 60.0%-137.8% with the relative standard deviations less than 16.0%.The limits of detection were 0.1μg/kg.Conclusion The developed method is selective,sensitive,simple,accurate and can be used to quantify the 15 kinds of PAHs controlled by European Union with priority in fruits,vegetables and cereals.
