重水标记法测定大鼠血浆IgG合成动力学相关参数
Deuterated water labelling-based determination of kinetic parameters of plasma immunoglobulin G synthesis in rats
【摘要】 为测定大鼠血浆免疫球蛋白G(IgG)合成动力学相关参数,以重水(2 H2O)为示踪剂,将20只SD大鼠以腹腔注射、日常饮水的形式进行2 H2O标记,分别于首次标记后第1、3、5、6、10周末,各取4只大鼠,心脏穿刺采集血样,采用蛋白纯化仪纯化大鼠血液中IgG,并用聚丙烯酰胺凝胶电泳(SDS-PAGE)鉴定,将提纯的IgG水解,分离纯化IgG中丙氨酸及血浆游离丙氨酸,并衍生,用气相色谱-质谱仪(GC-MS)检测血浆IgG中2 H-丙氨酰基丰度和血浆游离丙氨酸中2 H丰度;计算IgG分数合成率(FSR)、合成动力学方程。结果表明:血浆IgG的FSR为14.83%·周-1,合成动力学方程为ft=0.084 1×(1-e-0.148 3t)。这一研究提示2 H2O标记法能有效测定血浆IgG合成动力学参数,可用于长期标记试验。
【Abstract】 To determine the kinetic parameters of plasma immunoglobulin G(IgG)synthesis in rats by using deuterated water(2 H2O)labelling method,twenty SD rats were labeled with 2 H2O by intraperitoneally injection and drinking.At the end of the 1st,3rd,5th,6th and 10th week after the first labeling,four rats were sacrificed by cardiac puncture for blood collection.Plasma IgG was purified by protein chromatography,identified by polyacrylamide gel electrophoresis(SDS-PAGE)and hydrolyzed.The alanine in IgG and plasma was separated,purified and derived.The enrichment of 2 H-alanyl from IgG and the enrichment of 2 H from free alanine in plasma were measured by gas chromatography-mass spectrometry(GC-MS)respectively.The fractional synthesis rate of IgG and synthetic dynamic equation were further calculated.The results showed that the fractional synthesis rate of plasma IgG was 14.83%per week,synthetic dynamic equation was ft=0.084 1×(1-e-0.148 3t).2 H2O labeling method can effectively determine the kinetic parameters of plasma IgG synthesis and can be used for long-term labeling experiments.
