葛根汤颗粒与葛提取物对小鼠急性酒精性肝损伤的影响

【摘要】 目的研究葛根汤颗粒与葛提取物对小鼠急性酒精性肝损伤的保护作用,并探讨其可能机制,从而为临床应用提供依据。方法将60只雄性ICR小鼠随机分为正常组、模型组、联苯双酯滴丸组(阳性药组)、葛根汤颗粒组、葛根水提物组、葛花水提物组。除正常组外,其余各组每天灌胃52度二锅头酒,建立急性酒精性肝损伤模型;造模同时,灌胃给予受试药,每日1次,连续14d。末次给药后各组禁食不禁水,12h后摘眼球取血,测定血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)含量、胆固醇(TC)、甘油三酯(TG)含量、碱性磷脂酶活性(ALP)。测定肝组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果葛根汤颗粒能明显降低肝组织中MDA含量,降低血清中ALT、AST、TC、TG、ALP水平及肝脏系数,提高肝组织中SOD活性。葛根水提物能降低肝组织中MDA含量,降低血清中ALP水平及肝脏系数,提高肝组织中SOD活性。葛花水提物能降低肝组织中MDA含量和肝脏系数,提高肝组织中SOD活性。结论葛根汤颗粒对急性酒精性肝损伤具有一定的保护作用,其机制可能与抗氧化有关;葛提取物对急性酒精肝有不同程度的保护作用。

【Abstract】 Objective To study the protective effect of Puerariae decoction granules and extracts of Pueraria lobata on acute alcoholic liver injury in mice,elucidate the possible mechanism and to provide data for clinical use.Methods Sixty ICR male mice were randomly divided into six groups:normal group,model group,positive control group,Puerariae decoction granule group,water extracts of Pueraria lobata group,and water extracts of Pueraria lobata flowers group.Except the normal group,the other groups were administered with 52% Red Star Erguotou liquor,so that an acute alcoholic liver injury model was established.At the same time,each group was treated with drugs for 14 days except the normal control group and model group.After 12 hof the last administration,the blood was separated from eyeball,while levels of alanine transaminase(ALT),aspartate transaminase(AST),totalch olesterol(TC),triglyceride(TG)and alkaline phosphatase(ALP)in serum were detected.The activities of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in liver tissue were determined.Results Compared with the model group,Puerariae decoction granules could significantly reduce the content of MDA in liver tissue and the serum levels of ALT,AST,TC,TG,and ALP and the liver factor,while improving the SOD activity of liver tissue.Water extracts of Pueraria lobata could also reduce the content of MDA in liver tissue,the serum levels of ALT,AST and ALP and liver factor,and improve the SOD activity of liver tissue.Water extracts of Pueraria lobata flowers could reduce the content of MDA in liver tissue,the serum levels of ALT and AST and liver factor,and improve the SOD activity of liver tissue.Conclusion Puerariae decoction granules have protective effects on acute alcoholic liver injury.The mechanism may be related to antioxidation.Extracts of Pueraria lobata have protective effects on acute alcoholic liver injury to different extents.
