人低分化肺腺癌及癌旁组织的差异蛋白质组学研究

【摘要】 目的建立低分化腺癌及癌旁正常支气管上皮组织的差异蛋白,以寻找肺腺癌的潜在蛋白标记物。方法对6例低分化肺腺癌、癌旁正常组织进行蛋白质组学分析,即利用双向凝胶电泳的方法分离二者的蛋白质,用图像分析软件找出差异蛋白,应用基质辅助激光电离飞行时间质谱MALDI-TOF-MS得到肽质量指纹图谱,然后搜索数据库鉴定差异蛋白。结果建立人肺腺癌及癌旁正常组织的双向凝胶电泳图谱,低分化腺癌及癌旁正常组织的平均蛋白点数分别为(2066±144)、(1549±33)。对15个差异蛋白点肽质量指纹串联图谱鉴定,鉴定出一些与肿瘤转移、细胞周期调控、抑癌基因等蛋白质。结论 2-DE结合MALDI-TOF-MS/MS可鉴定肺腺癌及癌旁组织的蛋白,为进一步筛选肺腺癌的分子标志物、靶向治疗及预后的判断奠定了基础。

【Abstract】 Objective To identify potential proteinmarkers in lung adenocarcinomas. Methods Comparative proteome analysis of human poorly differentiated lung adenocarcinoma and paired normal bronchial mucosa adjacent to tumors and moderately differentiatedtissues from 6 cases were carried out. Total proteins of the carcinoma tissue and normal bronchialmucosa were seperated by means of immobilized Phgradient based twodimensional gel electrophoresis( 2-DE). The differentially expressed proteins wereanalyzed and identified by matrixassisted laser desorptionionization time of flight mass spectrometry( MALDI-TOF-MS/MS) and database searching. Results The average spots for human poorly differentiated lung adenocarcinoma and paired normal bronchial mucosa adjacent to tumors and moderately differentiatedtissues were( 2066 ± 144) 、( 1549 ± 33),respectively. The test were repeated for three times. 15 proteins were identified by peptide mass fingerprint( PMF). Some proteins were the products of the neoplasm,and the others are involved in cell cycle regulation,tumormetastasis,tumorsuppressor. Conclusion 2-DE and MALDI-TOF-MS/MS can identify proteins differentially expressed in the tumor. These proteins may potential biomakers of the lung adenocarcinoma and also provide further insights into the prognosis and treatment of the study on the cancer.
