葛根绞股蓝混合提取物对乙醇损伤大鼠肝脏的保护作用

【摘要】 目的研究葛根绞股蓝混合提取物对乙醇损伤大鼠肝脏的保护作用。方法乙醇经口灌胃建立大鼠亚急性肝损伤模型,以护肝片作为阳性对照组,受试药葛根绞股蓝混合提取物分为低、中、高(0. 33、0. 66、1. 00 g/kg)剂量组,灌胃30天。观察各组大鼠体重、自然状态情况,测定大鼠血清总胆红素、总胆固醇、低密度脂蛋白含量,测定肝脏中CYP2E1的表达,并对肝脏组织进行病理组织学检查。结果与模型组相比,葛根绞股蓝混合提取物组亚急性肝损伤大鼠的血清中总胆红素、总胆固醇、低密度脂蛋白含量显著降低。肝脏中乙醇诱导的CYP2E1表达降低。病理组织学检查可见葛根绞股蓝混合提取物中、高剂量组肝细胞脂肪变性较模型组均有减小,坏死程度下降。结论葛根绞股蓝混合提取物具有保护肝脏的作用,且其作用强度与护肝片无明显差异。

【Abstract】 Objective To evaluate the effect of Puerariae Radix and Gynostemma pentaphyllum. Makino extract( i. e. PR&GP extract) on alcohol induced rat liver injury. Methods Subacute rat alcoholic injury model was established by intragastric administration of ethanol.KuiHua Hugan tablet was used as positive control,and low,medium and high( 0. 33,0. 66,1. 00 g/kg) dose of PR&GP extract were orally given to rats for 30 days. The states of rats were observed and recorded. The serum total bilirubin( TBIL),total cholesterol( TC) and low-density lipoprotein( LDL) content were measured. Rat liver was taken out to carry out histologic examination and expression of cytochrome P450 2 E1 was tested. Results The PR&GP extract significantly reduced serum TBIL,TC and LDL of ALD Rats,reduced ethanol-induced CYP2 E1 expression in rat liver,and histologic analysis showed that PR&GP extract of middle and large dose reduced the steatosis of hepatocyte and degree of necrosis of the hepatic cord compared with model group. Conclusion PR&GP extract has protection of hepatocytes in ALD rats but no significant difference compared with KuiHua Hugan tablet.
