载阿奇霉素CS/PLGA缓释材料的制备与性能研究
Preparation and properties research of CS/PLGA slow-release composites loading azithromycin
【摘要】 以壳聚糖(CS)、乳酸一羟基乙酸共聚物(PLGA)、阿奇霉素片为原料制备多孔载阿奇霉素CS/PLGA缓释材料。采用比表面及孔径分析仪(BET)、场发射扫描电子显微镜(FESEM)、热重-质谱联用仪(TG-MS)、紫外分光光度计(UV)等方法对多孔材料进行表征。BET结果表明所制的多孔材料平均孔直径均在2~50nm范围内,符合多孔材料的要求; FESEM结果表明多孔材料载药成功; TG-MS热分析结果表明,在500℃时,载阿奇霉素多孔材料的质量残留率为33. 8%,阿奇霉素的质量残留率为14. 2%,载阿奇霉素多孔材料热性能优于阿奇霉素药物本身; UV实验测定结果为该缓释剂的载药量为40. 63%,包封率为54. 03%,106h时累计释放率达到91. 72%,此后基本不再释放,缓释效果良好。
【Abstract】 The chitosan,PLGA and azithromycin were used as source materials,the CS/PLGA slow-release composites loading azithromycin were prepared.The properties were tested by specific surface area and pore size analysis( BET),field emission scanning electron microcopy( FESEM),thermogravimetry-mass spectrometry( TG-MS) and ultraviolet-visible spectrophotometry( UV). The results of BET proved that the average pore size of the composites were in the range of 2 ~ 50 nm,which satisfied the requirement of the porous composites; the results of FESEM proved that composites which loading azithromycin loaded successfully.The results of TGMS showed that when temperature came to 500℃,the residual mass of CS/PLGA composites loading azithromycin was 33. 8%,the residual mass of azithromycin was 14. 2%,the thermal properties of the CS/PLGA composites loading azithromycin was better than the azithromycin.The UV experiments determinated that the drug loading of reliever was 40. 63%,envelop rate was 54. 03%. It didn’t release when the release rate reached 91. 72%after 106 hours,which proved that the sustained-release effect of this reliever was the best.
