姜黄素吡唑衍生物与牛血清白蛋白的相互作用研究

【摘要】 在模拟生理酸度的环境下,用荧光法研究了姜黄素吡唑衍生物3,5-二(4-羟基-3-甲氧基苯基乙烯基)-N-脒基吡唑与牛血清白蛋白(BSA)的相互作用。实验结果表明:该合成化合物与牛血清白蛋白反应为静态猝灭,且荧光光谱有较大的红移;根据Stern—Volmer荧光猝灭方程计算得到该衍生物和BSA在298K和310K温度下的结合常数和结合位点数;由实验数据计算得到该猝灭反应的热力学参数△G<0,△H>0,△S>0,表明该衍生物与BSA之间相互作用为自发、吸热且以疏水作用力为主。

【Abstract】 In this paper,in the environment of physiological acidity,the fluorescence method was used to study the interaction between the derivative of pyrazole,3,5-bis( 4-hydroxyl-3-methoxyphenylvinyl)-N-amidine and bovine serum albumin( BSA). The experimental results show that the chemical reaction between synthesized compound and bovine serum albumin is static quenching,and fluorescence spectrum has a large red shift. According to the Stern-Volmer fluorescence quenching equation,binding constants and binding sites number of the derivative and BSA are calculated at 298 K and 310 K.According to Experimental data,the thermodynamic parameters of the quenching reaction are △G < 0,△H > 0 and △S > 0,indicating that the interaction between the derivative and BSA is spontaneous,endothermic,mainly based on hydrophobic force.
