凝集素修饰的Fe2O3@Au纳米粒子与SW620细胞的相互作用研究
Interactions of lectin-modified Fe2O3@Au nanoparticle with SW620 cells
【摘要】 首先采用共沉淀法及交互盐酸羟胺还原法制备Fe2O3@Au核壳结构纳米粒子,利用凝集素修饰纳米粒子(Lectin-Fe2O3@Au NP),然后通过动态光散射(DLS)/聚丙烯酰胺凝胶电泳/磁滞回曲线进行表征。利用MTT法测纳米粒子的细胞毒性,将不同浓度的Lectin-Fe2O3@Au与结直肠癌SW620细胞相互作用,普鲁士蓝进行染色,使用紫外可见光谱法(UV-Vis)和光学显微镜进行观察。结果表明凝集素修饰的纳米粒子在细胞培养基中能够稳定存在,当纳米粒子浓度为0.72 n M时,结直肠癌SW620细胞的存活率仍高于90%,其作用强度依次为蓖麻凝集素(RCA)-Fe2O3@Au>麦胚凝集素(WGA)-Fe2O3@Au>伴刀豆素(Con A)-Fe2O3@Au,经过普鲁士蓝染色的细胞可以观察到RCA-Fe2O3@Au纳米粒子的存在,表明凝集素RCA具有作为修饰纳米粒子与结直肠癌SW620细胞靶向识别的潜力。
【Abstract】 Fe2O3@ Au core-shell nanoparticles were successfully prepared by co precipitation method and the interaction of hydroxylamine hydrochloride. Further nanoparticles were modified by lectin,the structure and physicochemical characterizations were proved by dynamic light scattering( DLS)/polyacrylamide gel electrophoresis/magnetization curve.The cytotoxicity of the lectin-conjugated Fe2O3@ Au nanoparticles was evaluated through MTT cell proliferation assays,and dyed by Prussian blue.the interactions of three lectin-conjugated Fe2O3@ Au nanoparticles with colon cancer cell line SW620 were studied by optical ultraviolet-visible spectrometer and microscope.The results showed Lectin-Fe2O3@ Au nanoparticles had highly salt stability and stable in cell culture medium.The survival rate of colorectal cancer SW620 cells was still higher than 90%when the concentration of nanoparticles was 0. 72 n M,the binding affinity with SW620 was( Ricinus Communis Agglutinin) RCA-Fe2O3@ Au>( Wheat Germ Agglutinin) WGA-Fe2O3@ Au>( Concanavalin) Con A-Fe2O3@ Au,the existence of RCA-Fe2O3@ Au nanoparticles could be observed in the cells which were dyed by the Prussian blue.These findings suggested that the RCA-Fe2O3@ Au could be potentially employed as a target agent in the recognition of colon cancer cell SW620.
