含香豆素结构的姜黄素衍生物的合成及生物活性

【摘要】 以单羰基姜黄素衍生物1,4-戊二烯-3-酮为先导,将具有良好生物活性的肟醚基团和香豆素基团引入其结构中,合成了6个新型姜黄素衍生物,其结构均通过1H NMR、13C NMR、IR及ESI-MS等方法进行了表征。生物活性测试结果表明:在药剂质量浓度为500 ug·m L-1时,大部分目标化合物对TMV均有一定的治疗和保护活性。其中,目标化合物3c、3d、3e和3f对TMV拥有较好的治疗作用,其抑制率分别为38.9、38.8、42.5和38.2%,略优于对照药剂病毒唑(36.2%)。此外,在药剂质量浓度为100和50 ug·m L-1时,化合物对水稻白叶枯菌和烟草青枯菌具有一定抑制活性,但相对较差。该项研究表明此类化合物对TMV拥有较好的抑制作用,在其结构基础上,进行更深一步的优化和改进,有望得到新型的抗植物病毒剂。

【Abstract】 Using curcumin as the lead molecule,six novel 1,4-penta-1,4-diene-3-one derivatives,which were confirmed by IR,1 HNMR,and 13 C NMR and ESI-MS,were synthesized by combing oxime ether and coumarin groups into one molecule.The antiviral bioassy at 500 ug·m L-1 indicated that some title compounds showed significant antiviral activity against tobacco mosaic virus( TMV),of which,the title compounds 3 c,3 d,3 e and 3 f exhibited better curative activities against TMV,with the inhibitory rates at38. 9,38. 8,42. 5 and 38. 2%,respectively,which were better than that of ribavirin( 36. 2%). Meanwhile,the antibacterial bioassys at 100 and 50 ug·m L-1 indicated that some title compounds showed significant antibacterial activities against Xanthomonas oryzae pv.oryzae and Pseudomonas solanacearum. These bioassys indicated that some compounds with potent antiviral activities might be obtained via the structural modification based on the penta-1,4-diene-3-one oxime derivatives bearing coumarin moiety.
