白花蛇舌草不同极性部位抗血管生成及抗氧化活性研究

【摘要】 目的:研究白花蛇舌草不同极性部位的体外抗血管生成和抗氧化活性,以期为白花蛇舌草深入研究与开发提供理论依据。方法:使用石油醚、乙酸乙酯、正丁醇对95%乙醇提取物水溶液依次萃取,得到4种不同极性部位。人脐静脉内皮细胞(HUVEC)管腔形成实验初步观察各极性部位抗血管生成活性;以总还原力、DPPH·和·OH清除率为评价指标,考察各萃取物抗氧化能力。结果:石油醚、乙酸乙酯萃取物对HUVEC管腔形成有较好抑制作用;乙酸乙酯部位总还原力高于其他部位;DPPH·清除率顺序为:乙酸乙酯>正丁醇>石油醚>水部位,其IC50值分别为0.110 mg/m L、0.122 mg/m L、0.473 mg/m L和0.754 mg/m L,·OH清除率顺序为:石油醚>正丁醇>乙酸乙酯>水部位,其中石油醚部位清除·OH线性关系较好,线性方程为y=38.565x-40.991,IC50值为2.374 mg/m L。不同极性部位对HUVEC管腔形成的抑制和抗氧化活性呈浓度依赖关系,DPPH·清除率呈时间依赖关系。结论:白花蛇舌草不同极性部位具有一定抗氧化活性,同时能抑制HUVEC管腔形成。

【Abstract】 Objective: To study the anti-angiogenesis and antioxidant activity of different polarity fractions of Baihuasheshecao(Oldenlandiae) in vitro and to provide theoretical basis for the follow-up research and development of Baihuasheshecao(Oldenlandiae). Methods: Petroleum ether, ethyl acetate and n-butanol were used to extract the 95% ethanol extract in turn, and four different polar fractions were obtained. Human umbilical vein endothelial cells(HUVEC) were used to study the anti-angiogenic activity of each extract. The antioxidant capacity of each extract was investigated by using the total reducing power, DPPH· and OH· scavenge rate as evaluation indexes. Results: The formation of HUVEC lumen was inhibited better by petroleum ether and ethyl acetate fraction. The total reducing power of ethyl acetate fraction was higher than other fractions. The order of DPPH· free radicals scavenge rate was: ethyl acetate>n-butanol>petroleum ether>water fraction, while IC50 values were 0.110 mg/m L, 0.122 mg/m L, 0.473 mg/m L and 0.754 mg/m L in turn. The order of hydroxyl radical(·OH)scavenge rate was: petroleum ether >n-butanol >ethyl acetate >water fraction, the linear relationship of petroleum ether fraction scavenging ·OH was better and the linear equation was y=38.565 x-40.991 and the IC50 value was2.374 mg/m L. The inhibitory effect of different polarity fractions on the formation of HUVEC lumens was in a concentration-dependent manner, and the DPPH· scavenging rate showed time-dependent relationship. Conclusion: It was found that the extracts of Baihuasheshecao(Oldenlandiae) had some antioxidant activity and could inhibit the formation of HUVEC lumen.
