白黎芦醇对宫颈癌Hela细胞中人程序化死亡分子5蛋白、mRNA表达的影响
Effects of Resveratrol on the Protein and mRNA Expressions of Human Programmed Cell Death 5 in the Cervical Carcinoma Hela Cells
【摘要】 目的探讨白黎芦醇对宫颈癌Hela细胞凋亡相关基因人程序化死亡分子5(PDCD5)蛋白、mRNA表达的影响。方法将宫颈癌Hela细胞随机分为空白对照组及4个浓度的白黎芦醇组[分别用25、50、100、200μmol·L-1白藜芦醇处理宫颈癌Hela细胞24、48、72 h],运用免疫细胞化学、原位杂交技术分别检测处理前后宫颈癌Hela细胞中PDCD5蛋白、mRNA的表达。应用TUNEL法检测各组细胞凋亡的情况。结果 4种不同浓度的白黎芦醇分别处理宫颈癌Hela细胞24、48、72 h后,与空白对照组比较,PDCD5蛋白、mRNA的表达均有明显升高,且白藜芦醇的浓度越高,其表达也越高,差异均有统计学意义(P均<0.05);同一浓度白藜芦醇处理组中,随着时间延长,PDCD5蛋白、mRNA的表达也均升高,差异均有统计学意义(P均<0.05)。4种不同浓度的白黎芦醇分别处理宫颈癌Hela细胞24、48、72 h后,与空白对照组比较,细胞凋亡指数明显增加,且随白藜芦醇的浓度的增加而升高,差异有统计学意义(P<0.05);但同一浓度白藜芦醇处理组细胞凋亡指数随白藜芦醇的作用时间延长增加均不显著,差异均无统计学意义(P均>0.05)。结论 PDCD5蛋白在宫颈癌Hela细胞中的表达可能与宫颈癌的发生、发展过程相关,白黎芦醇可上调宫颈癌Hela细胞中PDCD5蛋白、mRNA的表达,进而诱导宫颈癌Hela细胞的凋亡。
【Abstract】 Objective To explore the effects of resveratrol on human programmed cell death 5(PDCD5) expression in the cervical carcinoma Hela cells.Methods Cervical carcinoma Hela cells were randomly divided into the blank control group and the 4 doses of resveratrol groups[25, 50, 100 and 200 μmol·L-1 resveratrol for cervical carcinoma Hela cells 24, 48, 72 h], The expressions of PDCD5 protein and mRNA in the cervical carcinoma Hela cells before and after treatment were detected by immunocytochemistry and in situ hybridization.TUNEL method was used to detect cell apoptosis in each group.Results Compared with the blank control group, the expressions of PDCD5 protein and mRNA were significantly increased of the cervical carcinoma Hela cells in the 4 doses of resveratrol group treated 24, 48 and 72 hours with different concentrations of resveratrol, and the higher the concentration of resveratrol, the higher the expression level, and the difference was statistically significant(P<0.05).In the same concentration group, the expression of PDCD5 protein and mRNA were increased with time(P<0.05).Compared with the blank control group, the apoptosis index were significantly increased of the cervical carcinoma Hela cells in the 4 doses of resveratrol group treated 24, 48 and 72 hours with different concentrations of resveratrol, and the higher the concentration of resveratrol, the higher the apoptosis index, and the difference was statistically significant(P<0.05).In the same concentration group, the expression of PDCD5 protein and mRNA were increased with time(P<0.05).But the increase of apoptosis index in the same concentration of resveratrol was not significant(P>0.05).Conclusion The expression of PDCD5 in the cervical carcinoma Hela cells may be related to the development of cervical carcinoma, resveratrol could up-regulate Hela cell apoptosis through PDCD5 protein and mRNA in the cervical carcinoma Hela cells.
