颗粒物与花粉联合作用对细胞氧化损伤的影响

【摘要】 目的探讨不同颗粒物与致敏花粉联合作用对细胞的氧化损伤。方法采用200μg/ml发电厂燃煤飞灰、豚草花粉和豚草花粉+燃煤飞灰混和物染毒中国仓鼠肺细胞(CHL),检测细胞丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力、谷胱甘肽过氧化物酶(GSH-PX)活力,并观察0.01 mol/L N-乙酰半胱氨酸(NAC)抗氧化效应。结果与对照组比较,花粉、燃煤飞灰和混合物染毒组的MDA含量均升高,T-SOD活力均降低(P<0.05);GSH-PX活力均呈现下降趋势,但仅混合物染毒组差异有统计学意义(P<0.05)。与豚草花粉或燃煤飞灰单独染毒比较,混合物染毒组的MDA含量升高(P<0.05),T-SOD、GSH-PX活力虽呈现下降趋势,但未见统计学意义(P>0.05)。与混合物染毒组比较,混合物+NAC组MDA含量降低,T-SOD活力升高(P<0.05)。结论燃煤飞灰和豚草花粉均可造成CHL细胞的氧化损伤,并且当豚草花粉和燃煤飞灰联合作用时,其氧化损伤作用有增强的趋势,抗氧化剂可以对花粉和颗粒物的氧化损伤起到拮抗作用。

【Abstract】 Objective To study the combined effect of particulates and allergic pollen on oxidative damage in CHL cells.Methods Chinese hamster lung cells(CHL) were exposed to 200 μg/ml coal-fired fly ash from a power plant, ragweed pollen,and their mixture, the levels of malondialdehyde(MDA), superoxide dismutase(SOD) activity and glutathion peroxidase(GSH-Px)activity were determined, as well as the antioxidative effects of 0.01 mol/L NAC. Results Compared with the control group,higher content of MDA and lower activity of T-SOD were observed in fly ash alone exposure group and pollens alone exposure groups(P<0.05), lower activity of GSH-Px was olny observed in mixture exposure group(P<0.05). Compared with the fly ash alone exposure group or ragweed pollen alone exposure group, mixture exposure group showed higher content of MDA(P<0.05),lower activities of T-SOD and GSH-Px(P>0.05). Compared with the mixture exposure group,lower content of MDA and higher activity of T-SOD were observed in mixture+NAC intervention group(P<0.05). Conclusion Coal-fired fly ash and ragweed pollen both can cause oxidative damage in CHL cells, furthermore, their combined effect may increase oxidative damage,which can be antagonized by antioxidants.
