miRNA-221和miRNA-222与甲状腺癌侵袭转移的相关性研究

【摘要】 目的探讨miRNA-221和miRNA-222与甲状腺癌侵袭转移的相关性。方法选择2013年10月至2016年10月接受甲状腺次全手术切除治疗的病理确诊的甲状腺癌患者168例作为研究对象,于确诊后收集患者外周静脉血与新鲜标本提取miRNA,进行RT-PCR检测血浆及组织miRNA-221及miRNA-222表达水平。结果癌组织miRNA-221及miRNA-222与血浆miRNA-221及miRNA-222表达呈正相关(r=0. 226,P <0. 05);血浆miRNA-221及miRNA-222表达水平在不同年龄、淋巴结转移与否、腺体外浸润与否、肿瘤大小及TNM分期均有差异,且有统计学意义(P <0. 05)。结论 miRNA-221及miRNA-222在甲状腺癌组织及血浆中高表达,与甲状腺癌侵袭转移的明显相关,可能可以作为生物标志物。

【Abstract】 Objective To investigate the correlation of miRNA-221,miRNA-222 and invasion and metastasis of thyroid cancer. Methods A total of 168 patients with thyroid cancer who were pathologically confirmed as thyroid cancer and underwent subtotal thyroidectomy in our hospital from October 2013 to October 2016 were enrolled in this study. After diagnosis,the peripheral venous blood and fresh specimens of patients were collected for miRNA extraction. And RT-PCR was used to detect expression levels of miRNA 221 and miRNA 222 in the plasma and tissues. Results The expression levels of miRNA 221 and miRNA 222 in cancerous tissues were positively correlated with those in the plasma( r = 0. 226,P < 0. 05).There were significant differences in the expression levels of plasma miRNA 221 and miRNA 222 in terms of age,presence of lymph node metastasis and in vitro infiltration,tumor size and TNM staging( P < 0. 05). Conclusion The miRNA-221 and miRNA-222 are highly expressed in the tissues and plasma of patients with thyroid cancer,which are closely correlated with invasion and metastasis of thyroid cancer. Therefore,it may be used as biomarkers.
