转化生长因子β1与三阴性乳腺癌临床特征的关系及信号通路研究
Correlation between transforming growth factor beta 1 and three negative breast carcinoma
【摘要】 目的通过研究三阴性乳腺癌组织中转化生长因子β1表达情况,阐明TGF-β1的表达强度与三阴性乳腺癌患者各项临床病理特征、预后之间的关系,并初步探讨TGF-β1对三阴性乳腺癌细胞的生物学效应及可能的信号传导通路。方法采用免疫组化法研究三阴性乳腺癌组织中TGF-β1表达阳性率的变化,与非三阴型组对照,分析其与三阴型乳腺癌临床病理特征及预后之间的关系,同时采用TGF-β1处理MDA-MB-231,测定其处理前后侵袭迁移能力的变化及信号通路蛋白的变化。结果在三阴型乳腺癌患者癌灶组织中TGF-β1的高表达率显著高于非三阴组(P<0.05),并且通过5年的随访研究,TGF-β1的表达在三阴性乳腺癌患者中,年龄、绝经状态、肿瘤家族史、肿瘤大小等方面无显著差异(P>0.05),肿瘤的组织学分级和腋下淋巴结转移则呈显著相关(P<0.05),其高表达率越高,五年无瘤生存率越低,TGF-β1能够促进MDA-MB-231细胞的侵袭转移,且Smad2蛋白和P38蛋白表达增高。结论三阴性乳腺癌组织中TGF-β1表达高表达率与三阴性乳腺癌组织学分级和腋下淋巴结是否转移呈显著相关,并影响三阴性乳腺癌患者的5年无瘤生存率,TGF-β1能够能够增强MDA-MB-231细胞的侵袭迁移能力,Smad2蛋白通路和P38信号通路可能发挥着重要的作用。
【Abstract】 Objective To investigate the expression of transforming growth factor beta 1( TGF-β1) in three negative breast cancer( TNBC) tissues to explore the correlation between the expression levels of TGF-beta 1 and clinical pathological characteristics as well as prognosis of TNBC,and to explore the biological effects of TGF-β1 on three negative breast cancer cell and the possible signal transduction pathway. Methods The immunohistochemical method was used to investigate the changes of positive rate of TGF-β1 in TNBC,and to compare them with those in non three negative breast cancer group,at the same time,TGF-β1 was used to treat MDA-MB-231,then the changes of invasion and migration ability before and after treatment,and the protein changes in signaling pathways were observed. Results The positive expression rates of TGF-β1 in three negative breast cancer tissues were significantly higher than those in non three negative breast cancer group( P < 0. 05),moreover,there were no significant differences in expression levels of TGF-β1 in different ages,menopausal status,family history of cancer,tumor size,however,which were closely correlated with tumor histological grade and axillary lymph node metastasis( P < 0. 05),the higher the expression levels,the lower the 5-year survical rate. In addition TGF-β1 could promote the invasion and migration of MDA-MB-231,and the expression levels of Smad2 protein and P38 protein were increased.Conclusion The high expression rate of of TGF-β1 in TNBC tissues is closely correlated to tumor histological grade and axillary lymph node metastasis,which affects 5-year disease-free survival rate of patients with TNBC,moreover,TGF-β1 can enhance the invasion and migration of MDA-MB-231 cells,and Smad 2 protein and P38 signaling pathway may play an important role in the process.
