三种生菜离体快繁的初步研究
Preliminary Study on Rapid Propagation in Vitro of Three Kinds of Lactuca sativa
【摘要】 以3种生菜子叶为外植体,研究了不同植物生长调节剂对生菜(Lactuca sativa)不定芽的诱导和植株离体再生的影响。结果表明,意大利生菜、玻璃生菜和结球生菜不定芽诱导最适培养基分别为MS+0.1mg/L 6-BA+0.2 mg/L NAA、MS+0.1 mg/L 6-BA+0.1 mg/L NAA和MS+0.2 mg/L 6-BA+0.1 mg/L NAA;3种生菜生根最适培养基分别为1/2 MS+0.05 mg/L NAA、1/2 MS和1/2 MS+0.1 mg/L NAA。通过分析影响3种生菜高频离体再生体系建立的因素,为进一步利用基因工程技术开发功能性蔬菜奠定了基础。
【Abstract】 In this experiment,three kinds of cotyledons from Lactuca sativa were used as explants to study the effects of different plant growth substances on the induction of adventitious buds and the in vitro regeneration. The results showed that the optimal medium for the induction of adventitious buds from Lactuca sativa L. var. ramosa Hort.,Lactuca sativa L. var. crisha Hort. and Lactuca sativa var capitata L. were MS +0.1 mg/L 6-BA +0.2 mg/L NAA,MS +0.1 mg/L 6-BA +0.1 mg/L NAA,and MS+0.2 mg/L 6-BA+0.1 mg/L NAA,respectively. The optimum rooting medium for the three kinds of Lactuca sativa were 1/2 MS+0.05 mg/L NAA,1/2 MS,and 1/2 MS+0.1 mg/L NAA. By analyzing the factors that affect the establishment of high-frequency regeneration system of three kinds of Lactuca sativa,it laid a foundation for further development of functional vegetables by using genetic engineering technology.
