二甲双胍对大鼠肝脏缺血再灌注损伤抗氧化保护作用
Antioxidative Effect of Metformin on Hepatic Ischemia-Reperfusion Injury in Rats
【摘要】 目的本文探讨二甲双胍对大鼠肝脏缺血再灌注损伤是否具有抗氧化保护作用。方法 30只SD大鼠随机分为5组,假手术组,溶剂组,二甲双胍高(200mg·kg-1)、中(100mg·kg-1)、低(50mg·kg-1)剂量组。观察各组肝脏生化指标及组织形态学变化。结果再灌注4h后,与溶剂组相对比,二甲双胍实验组的血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、肝组织匀浆丙二醛(MDA)的水平均降低,肝脏细胞损伤减轻,其中MDA在中剂量组降低的程度最为显著(P<0.05)。结论二甲双胍可减轻肝脏缺血再灌注损伤,并且具有抗氧化作用。
【Abstract】 OBJECTIVE To investigate the antioxidative effect of metformin on hepatic ischemia-reperfusion injury in rats. METHODS 30 SD rats were randomly divided into 5 groups,including sham operation group,solvent group,metforminhigh-dose group( 200 mg · kg-1),middle-dose group( 100 mg·kg-1),low-dose group( 50 mg·kg-1). The changes of liver biochemical markers and pathology were assessed. RESULTS After reperfusion for 4 h,compared with the solvent group,in the metformin group,the contents of alanine aminotransferase( ALT),aspartate aminotransferase( AST) in the serum,malondialdehyde( MDA) in the liver tissue and the liver cells damage was decreased. The content of MDA in the middle-dose group was lower than the other metformin groups( P < 0. 05). CONCLUSION Metformin can alleviate hepatic ischemia-reperfusion injury,and has antioxidative effect on liver cells in rats.
