白藜芦醇抑制缺氧诱导的肝癌细胞株HepG2侵袭并降低CXCR4表达
Resveratrol Inhibits Hypoxia Induced Invasion of HepG2 Cells and Decreased the Expression of CXCR4
【摘要】 目的探讨不同浓度白藜芦醇在缺氧环境下对肝癌细胞株HepG2侵袭的影响并探讨其机制。方法用不同浓度的白藜芦醇作用于人肝癌细胞株HepG2,用Transwell实验检测缺氧条件(Co Cl2)下白藜芦醇对肝癌细胞侵袭转移的影响,Western blot方法检测趋化因子受体CXCR4表达水平的变化,CXCR4特异性抑制剂AMD3100孵育HepG2细胞,观察其侵袭转移活性的变化。结果与对照组比较,白藜芦醇可以浓度依赖性的抑制缺氧条件下HepG2细胞的侵袭转移,Western blot结果提示白藜芦醇可降低CXCR4蛋白表达,AMD3100孵育能显著抑制HepG2细胞侵袭转移。结论本文揭示了白藜芦醇可抑制缺氧条件下人肝癌细胞株HepG2的侵袭转移,其抑制机制可能与白藜芦醇降低CXCR4蛋白表达水平有关。
【Abstract】 OBJECTIVE To investigate the effect of different concentrations of Resveratrol on invasion of HepG2 cells and to explore its mechanism. METHODS The effects of Resveratrol on the invasion and metastasis of HepG2 cells under hypoxia conditions were detected by transwell test; Western blot was used to detect CXCR4 expression; the effects of AMD3100 on the invasion and metastasis of HepG2 cells was observed by transwell test. RESULTS Compared with the control group,Resveratrol could inhibit the invasion and metastasis of HepG2 cells in a concentration dependent manner,and Western blot showed that Resveratrol could reduce CXCR4 protein expression; AMD3100 incubation Significantly reduced the invasion and metastasis of HepG2 cells. CONCLUSION Resveratrol can inhibit the invasion and metastasis of HepG2 cells under hypoxia conditions,the mechanism may be involved the decrease of CXCR4 protein expression by Resveratrol.
