UPLC-MS/MS法同时测定党参中7种核苷的含量
Simultaneous determination of seven nucleosides in Codonopsis pilosula by UPLC-MS/MS
【摘要】 目的建立超高效液相色谱-电喷雾四极杆质谱同时测定党参中7种核苷类成分含量的方法。方法采用Acquity UPLC BEH C18柱(2.1mm×50mm,1.7μm);以甲醇(A)-0.1%甲酸水(B)为流动相进行梯度洗脱,流速为0.2mL/min;进样量2μL;柱温30℃;电喷雾离子源(正离子模式),多反应监测模式(MRM)进行含量测定。结果 7种核苷类成分分别在1.8380.31、0.2084.03、0.2114.12、0.2134.07、0.2060.412、0.0070.913、0.0080.843 mg/L浓度范围线性良好,平均回收率为95.56%104.03%(RSD<3%)。结论所建立的分析方法快速、简便、灵敏、稳定性好,可用于党参饮片中7种核苷类成分含量的同时测定。
【Abstract】 Objective To establish a ultra performance liquid chromatographytandem mass spectrometry(UPLCMS/MS)method for the simultaneous determination of seven nucleosides in Codonopsis pilosula. Methods The determination was performed on the Acquity UPLC BEH C18 column(2.1 mm × 50 mm, 1.7μm), the methyl alcohol(A)-0.1% formicacid aqueous(B) in gradient elution as mobile phase. The flow rate was 0.2 m L/min, the sampling amount was 2μL, the column temperature was 30℃. The seven nucleosides were detected in a electrospray ionization mode(positive ion mode) and multiple reaction monitoring(MRM). Results Good linearity was achieved for quantitation in the range of 1.83-80.31, 0.208-4.03, 0.211-4.12, 0.213-4.07, 0.206-0.412, 0.007-0.913, 0.008-0.843 mg/L for seven nucleosides respectively. The average recovery rates were in the range of 95.56%-104.03% with RSD < 3%. Conclusion The developed method is rapid, simple sensitive, accurate, and highly repeatable which can be used for qualitative and quantitative analysis for nucleosides in Codonopsis pilosula.
