雷帕霉素抗阿尔兹海默病的机制研究
Review on the Mechanisms of Rapamycin Against Alzhermer’s Disease
【摘要】 大环内酯类抗生素雷帕霉素主要作为免疫抑制剂用于器官移植,近年来亦有许多研究表明它可通过抑制哺乳动物雷帕霉素靶标蛋白(m TOR)、促进自噬等途径发挥神经保护功能。本文就雷帕霉素抗阿尔兹海默病的分子机制研究进展进行综述,主要介绍其在抗衰老、减轻认知损伤、减少β-淀粉样蛋白(Aβ)聚集、促进Aβ的清除、减少Tau蛋白过度磷酸化、抑制NO大量产生以及抗阿尔兹海默的其他神经保护作用等方面的作用。
【Abstract】 Macrolide antibiotic rapamycin was mainly as an immunosuppressant for organ transplantation. Recently many studies have shown that through inhibiting the mammalian target of rapamycin protein(m TOR), rapamycin could promoted autophagy and other ways to play a neuroprotection function. The research advanced about the molecular mechanisms of rapamycin against Alzheimer’s disease was carried out summarized, introducing its effects about anti-aging, alleviating cognitive impairment, reducing Aβ aggregation, promoting the removal of Aβ,decreasing hyperphosphorylation of Tau,suppressing a large number of NO generated and neuroprotective effect of other aspects.
