银杏果壳多糖多酚提取及清除自由基活性研究
Extraction and Free Radical Scavenging Activity of Polysaccharide and Polyphenol from the Ginkgo biloba L. Shell
【摘要】 银杏果壳粉经石油醚脱脂后,用蒸馏水提取,通过单因素实验和正交实验确定银杏果壳多糖提取最佳工艺;同时研究银杏果壳多糖和多酚的体外清除自由基活性。结果表明,银杏果壳多糖最佳提取条件为:固液比1∶25(g:mL)、提取温度80℃、提取时间1h,提取率89.74%;多糖对DPPH、超氧阴离子自由基和ABTS自由基的清除能力相对较强;多酚对超氧阴离子自由基、DPPH和ABTS自由基的清除能力非常强,其IC50值分别为0.35、0.10和0.78g/L,表明银杏果壳多酚是一种较好的天然抗氧化剂。
【Abstract】 After de-fatted by petroleum ether,the residue of drying powder of ginkgo biloba L.shell was extracted using distilled water as the solvent.The extracting parameters,including solid-liquid ratio,extraction temperature and extraction time,were investigated with the designed single factor and orthogonal test method.The optimized extraction conditions were determined to be a solid to liquid ratio of 1∶25 with the extraction temperature at 80℃and an extracting duration of 1 h.The polysaccharide extraction rate under these conditions reaches 89.74%.Moreover,the scavenging capacities of polysaccharide and polyphenol from the ginkgo shell on DPPH,·OH,O-2·and ABTS+· were assayed.The results showed that the polysaccharide has scavenging effect on DPPH,O-2·and ABTS+·.The polyphenol has very strong scavenging effect on O-2·,DPPHand ABTS+·,with IC50 values of 0.35,0.10 and 0.78 g/L,respectively.It revealed that the polyphenol from the ginkgo shell is a promising natural antioxidants.
