2种牛角瓜离体叶片对低温胁迫的生理反应
Physiological Responses to Cold Stress in Detached Leaves of Two Kinds of Calotropis gigantea
【摘要】 分别将牛角瓜(Calotropis gigantea)、白花牛角瓜(Calotropis procera)成年植株的离体叶片置于不同的低温(15、10、5、0、-5、-10℃)条件下处理,以室温(20℃)为对照,测定分析叶绿素含量、可溶性蛋白质含量、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性等生理指标的变化。结果表明:随着胁迫温度的降低,叶绿素含量逐渐降低;可溶性蛋白含量表现为先升-后降-再升-再降的波浪形变化,且在-5℃时达到高峰;MDA含量总体先升后降,在15~5℃温度范围内,积累量最大;SOD表现为先降、后升、然后再降的曲线变化,而POD变化更复杂,但两种酶的活性基本都是在5℃上升至最高点。初步认为牛角瓜对低温胁迫具有一定的耐受能力,但其耐受力相对较差,一般5℃左右损伤已较大。
【Abstract】 The detached leaves of mature plants of Calotropis gigantea and Calotropis procera were treated under different conditions of low temperatures(15 ℃,10 ℃,5 ℃,0 ℃,-5 ℃ and-10 ℃) and 20 ℃(control). The changes of physiological indexes including chlorophyll content, soluble protein content,malondialdehyde(MDA) content,the activities of superoxide dismutase(SOD) and peroxide dismutase(POD) were studied. Results showed that the chlorophyll content declined gradually with the decrease ofstress temperature,and the soluble protein content showed a wave-shaped change of increasing at first and then decreasing and then increasing and then decreasing,and the peak came at-5 ℃;the content of MDA in-creased at first and then decreased,and the accumulation amount was the largest at the range of 15 ℃ to 5 ℃;the SOD showed a curve of decreasing at first,then increasing,and then decreasing again,and the change ofPOD was more complicated,but the activity of two enzymes raised to the highest point at 5 ℃. It was prelimi-narily considered that Calotropis gigantea had certain tolerance to low temperature stress,but its tolerancewas relatively poor,and the damage was generally serious at around 5 ℃.
