荧光光谱法测定淀粉中的支链淀粉

【摘要】 采用荧光光谱法测定了玉米淀粉中支链淀粉的含量。研究结果表明:支链淀粉在激发波长为392 nm、发射波长为453 nm处发射荧光;在0.02~0.10μg/mL浓度范围内,支链淀粉荧光强度与其浓度呈良好线性关系,相关系数r=0.9990,检出限为0.005μg/mL;用该方法测定玉米淀粉和土豆淀粉中支链淀粉的含量,加标回收率分别为98.74%和99.06%,相对标准偏差分别为2.53%和3.07%。该测定方法不需加任何生化试剂,不受时间、温度、共存直链淀粉的影响,快速简便、绿色环保、检出限低、结果准确。

【Abstract】 A method was developed for determination of amylopectin in corn starch by fluorescence spectrometry. The results showed that amylopectin had fluorescence at the excitation and emission wavelengths of 392 nm and 453 nm,respectively. The linear dynamic range was from 0.02 μg/mL to 0.10 μg/mL with the correlation coefficients of 0.9990,and the detection limit was 0.005 μg/mL. The method was applied to thedetermination of amylopectin content in corn starch and potato starch with recoveries 98.74% and 99.06%,respectively and the relative standard deviation was 2.53% and 3.07%,respectively. The method did not need addany biochemical reagent,and was not affected by time,temperature and coexistence of amylose. It was fast,simple,green and environmental friendly,with low detection limit and accurate results.
