莪术提取物榄香烯对肝癌H22荷瘤小鼠的抑瘤作用

【摘要】 目的研究莪术提取物榄香烯对肝癌H22荷瘤小鼠的抑瘤作用。方法选取昆明小鼠,用H22肝癌细胞悬液0.1 mL右腋下接种建立肝癌荷瘤小鼠模型。剩余17只作为正常组(右腋下注射0.9%Na Cl 0.1 mL)。将造模成功的小鼠按照体重随机分为3组,各17只:模型组、对照组、实验组。模型组和正常组腹腔内注射0.9%Na Cl 0.2 mL;对照组腹腔内注射氟尿嘧啶注射液20mg·kg-1;实验组腹腔内注射榄香烯注射液100 mg·kg-1,均qd,均连续给药10d。用称重法测定小鼠肿瘤重量并计算抑瘤率、胸腺指数和脾指数;用免疫组化法检测肿瘤组织生存素(Survivin)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)蛋白表达水平;用逆转录-聚合酶链反应检测肿瘤组织Survivin mRNA、Caspase-3 mRNA表达水平。结果给药后,模型组、对照组、实验组肿瘤重量分别为(3.09±0.43),(1.02±0.12),(1.24±0.15)g,对照组和实验组与模型组比较,差异均有统计学意义(均P<0.05)。对照组、实验组的抑瘤率分别为66.99%和59.87%,组间比较差异有统计学意义(P<0.05)。正常组、模型组、对照组、实验组肿瘤组织Survivin蛋白表达水平分别为(3.67±0.47)×104,(31.84.±2.74)×104,(23.84±2.75)×104,(27.85±3.05)×104,模型组与正常组比较,差异有统计学意义(P<0.05);对照组和实验组与模型组比较,差异均有统计学意义(均P<0.05)。与模型组比较,对照组、实验组的肿瘤组织Survivin mRNA表达水平较低,而Caspase-3蛋白、Caspase-3 mRNA表达水平较高,差异均有统计学意义(均P<0.05)。结论莪术提取物榄香烯能通过调节Survivin、Caspase-3表达来抑制肝癌H22荷瘤小鼠肿瘤生长。

【Abstract】 Objective To study the antitumor effect of elemene from Rhizoma Curcuma on hepatoma H22 tumor bearing mice. Methods Kunming mice were selected,51 liver tumor bearing mice models were established by right axillary inoculation with 0. 1 mL H22 hepatoma cell suspension,the remaining 17 mouse served as normal group and were injected normal saline 0. 1 mL into the right armpit. The mouse successfully modeled were randomly divided into model group,control group and experimental group,each with 17 mouse. The model group and the normal group were given sterile saline 0. 2 mL,qd,intraperitoneal injection; the control group was given fluorouracil injection 20 mg·kg-1,qd,intraperitoneal injection; the experimental group was given elemene injection 100 mg·kg-1,qd,intraperitoneal injection; mouse in each group were given continuous administration for 10 d. The tumor weight,tumor inhibitory rate,thymus index and spleen index were measured by weighing method. The levels of Survivin and Caspase-3 protein in tumor tissues were detected by immunohistochemistry. The expression levels of Surviving mRNA and Caspase-3 mRNA in tumor tissues were detected by reverse transcription polymerase chain reaction( RT-PCR).Results After administration,the tumor weight in model group,control group and experimental group were respectively( 3. 09 ± 0. 43),( 1. 02 ± 0. 12) and( 1. 24 ± 0. 15) g,comparison between control and experimental groups with the model group,the differences were statistically significant( all P < 0. 05). The tumor inhibition rates in the control group and the experimental group were respectively 66. 99% and 59. 87%,the differences between the two groups were statistically significant( P < 0. 05). The expression levels of Survivin protein in normal group,model group,control group and experimental group were( 3. 67 ± 0. 47) × 104,( 31. 84. ± 2. 74) × 104,( 23. 84 ± 2. 75) × 104,( 27. 85 ± 3. 05) ×104,comparison between normal group and model group,the differences were statistically significant( P < 0. 05); comparison between control and experimental groups with the model group,the differences were statistically significant( all P < 0. 05). Compared with the model group,the expression levels of Survivin mRNA in the tumor tissue of the control group and experimental group were lower,and the expression levels of Caspase-3 protein and Caspase-3 mRNA were higher,the differences were statistically significant( all P < 0. 05). Conclusion Elemene from Rhizoma Curcuma can inhibit tumor growth of hepatoma H22 tumor bearing mice by regulating the expression of Survivin and Caspase-3.
