苦参碱和氧化苦参碱对肝癌细胞增殖和凋亡的影响

【摘要】 目的研究苦参碱和氧化苦参碱对肝癌细胞增殖和凋亡的影响。方法将SMMC-7721肝癌细胞分为苦参碱组、氧化苦参碱组和对照组,苦参碱组用0.5,1.0,1.5,2.5,5.0,10.0 g·L-1的苦参碱干预,氧化苦参碱组用0.5,1.0,1.5,2.5,5.0,10.0 g·L-1的氧化苦参碱干预,对照组细胞无任何干预。用溴化四唑蓝(MTT)比色法检测SMMC-7721肝癌细胞的增殖情况,用双荧光染色法观察SMMC-7721肝癌细胞的凋亡情况。结果干预后4 d,10.0 g·L-1苦参碱组SMMC-7721肝癌细胞增殖抑制率为(85.37±4.82)%,高于对照组和10.0 g·L-1氧化苦参碱组的(4.16±3.92)%,(79.18±4.77)%(P<0.01);10.0 g·L-1苦参碱组的SMMC-7721肝癌细胞凋亡率为(69.85±4.41)%,高于对照组和10.0 g·L-1氧化苦参碱的(0.73±0.11)%,(59.18±4.08)%(P<0.01)。与对照组比较,苦参碱组和氧化苦参碱组各药物浓度下及不同培养时间下SMMC-7721肝癌细胞的增殖抑制率、凋亡率均显著升高(P<0.01),苦参碱组和氧化苦参碱组SMMC-7721肝癌细胞的增殖抑制率和凋亡率均随着培养时间和药物浓度的增加而增加(P<0.05或P<0.01),且相同浓度相同作用时间下,苦参碱组SMMC-7721肝癌细胞的增殖抑制率和凋亡率显著高于氧化苦参碱组(P<0.01)。结论苦参碱和氧化苦参碱均可增加SMMC-7721肝癌细胞增殖抑制率和凋亡率,且与作用时间、药物浓度呈正比,但苦参碱对SMMC-7721肝癌细胞的增殖抑制、促凋亡作用显著高于氧化苦参碱。

【Abstract】 Objective To investigate the effects of matrine and oxymatrine matrine on proliferation and apoptosis of hepatocellular carcinoma cells. Methods The SMMC-7721 hepatoma cells were divided into matrine group,oxymatrine matrine group and control group. Matrine group was intervened with 0. 5,1. 0,1. 5,2. 5,5. 0,10. 0 g·L-1 matrine,oxymatrine group was treated with 0. 5,1. 0,1. 5,2. 5,5. 0,10. 0 g·L-1 oxymatrine,control group was given no interference. Proliferation of SMMC-7721 cells was detected by Brominated four tetrazolium bluecolorimetric assay( MTT) method,double fluorescent staining was used to observe the apoptosis of SMMC-7721 cells in each groups. Results The inhibition rate of SMMC-7721 hepatocarcinoma cells after 4 d in10. 0 g·L-1 matrine group was( 85. 37 ± 4. 82) %,higher than those in control group and 10. 0 g·L-1 oxymatrine matrine group,which were( 4. 16 ± 3. 92) %,( 79. 18 ± 4. 77) %( P < 0. 01),The apoptosis rate of SMMC-7721 hepatocarcinoma cells after 4 d in 10. 0 g·L-1 matrinegroup was( 69. 85 ± 4. 41) %,higher than those in control group and 10. 0 g·L-1 oxymatrine matrine group,which were( 0. 73 ± 0. 11) %,( 59. 18 ± 4. 08) %( P < 0. 01). Compared with control group,the proliferation inhibition rate and apoptosis rate of SMMC-7721 cells were significantly increased under different drug concentrations and different culture time( P < 0. 01). The proliferation inhibition rate and apoptosis rate of SMMC-7721 cells were increased with the increase of culture time and drug concentration( P < 0. 05 or P < 0. 01). Under the same concentration,the inhibition rate and apoptosis rate of SMMC-7721 cells in matrine group were significantly higher than those in oxymatrine matrine group( P < 0. 01). Conclusion Matrine and oxymatrine matrine can increase the proliferation inhibition rate and apoptosis rate of SMMC-7721 hepatoma cells,and was proportional with the role of time,drug concentration,however,the inhibitory effect of matrine on the proliferation and apoptosis of SMMC-7721 hepatoma cells was significantly higher than that of oxymatrine matrine.
