卡瓦叶子的化学成分研究
Investigation on the Chemical Constituents from the Leaves of Kava
【摘要】 以卡瓦叶子粉末为研究对象,通过硅胶柱层析、薄层色谱法(TLC)跟踪、凝胶柱层析等方法对各个部位进行分离纯化,然后借助于核磁共振氢谱、碳谱、质谱等方法进行结构鉴定,最终从卡瓦叶子中分离出四个化合物,分别为:Dihydrokawain(1),5,7,4’-trimethoxyflavanone(2),芫花素(3), 5-hydroxy-7, 4’-dimethoxy-flavone(4)。采用MTT法,对四个化合物进行药理活性测试,结果发现,化合物1对细胞A549和细胞NCI-H460的IC50值分别为38.8136和38.5572μg/mL,有一定的细胞毒性,化合物2、3、4对细胞A549和NCI-H460没有明显的细胞毒性。
【Abstract】 Based on the investigation on the Chemical Constituents from the leaves of kava,four compounds were obtained from the EtO Acextract by a combination of column chromatography of silica gel,Sephadex LH-20 mamy times.Their structures were determined on the basis of 1HNMR,13C NMR,2DNMR,UV,IR,and ESIMS spectradata and were identified as as Dihydrokawain(1),5,7,4’-trime thoxy flavanone(2),genk wanin(3),5-hydroxy-7,4’-dimethoxy-flavone(4)The MTT assasy tested four compound scytotoxi city.the result showed that compound1 was found to be Moderate cytotoxicity against A549 and NCI-H460 Cells with IC50 values of 38.8136,38.5572μg/mL respectively.Compound2,3 and4 were found to be inactive toward all the tested cell A549 and NCI-H460(IC50>50μgmL-1).
