【摘要】 目的探讨一种利用鸡原始生殖细胞作为种子细胞,体外制备类胚体的方法。方法采用干细胞体外培养获得干细胞系并制备类胚体,采用免疫荧光染色、HE染色、RT-PCR及诱导分化等技术对干细胞系及类胚体的生物学特性进行鉴定分析。结果通过悬浮培养获得了最大体积为440μm的类胚体,并借助共聚焦显微镜活体细胞成像系统捕捉到其向心肌分化后自主收缩的影像。结论本研究提供了一种快速、高效的体外制备大体积类胚体的技术方法。更多还原

【Abstract】 Objective To prepare a novel method that preparing embryoid bodies by primordial germ cells in vitro.Methods The stem cell culture and inducing technologies were performed,the immunofluorescent staining,HE staining,RT-PCR and confocal laser scanning were utilized to identify the biological characteristics of primordial germ cells and embryoid bodies. Results Embryoid bodies with big size(maximum:440 μm in diameter)were obtained through the protocol.Conclusion This study provide a new technical platform to prepare embryoid bodies with big size in a short period with high efficiency.更多还原
